SRAP、TRAP、SSR、ISSR分子标记在水稻籼粳分型中的系统学研究
Molecular Phylogenetic Research on Classification of Indica and Japonica rice using SRAP, TRAP, SSR, ISSR markers
利用SRAP、TRAP新型分子标记,并结合SSR、ISSR标记对水稻47个材料进行了遗传多样性分析,四种分子标记多态性ISSR>SRAP>SSR>TRAP,SRAP标记的平均遗传相似性为0.70, TRAP标记0.74,SSR标记0.75,ISSR标记0.85;四种分子标记之间都呈极显著相关;聚类分析中籼粳稻都有较好的区分,在部分材料归类上有部分差异,但SRAP和TRAP较SSR和ISSR准确,而合并四种分子标记并聚类分析得到的树状图更能反映真正的系统演化关系。实验结果表明SRAP和TRAP标记在水稻研究都有较好的利用价值,SRAP标记在区分亲缘关系较近的品种间有更强的分辨能力。四种分子标记中爪哇稻Lemont,POO2都归为籼类群,说明它们与籼稻更近的亲缘关系。而普通野生稻材料在不同分子标记的树状图中,部分材料在籼粳归类上存在差异,在合并四种标记聚类分析中,所有野生稻材料都归为粳稻一类群,表明中国普通稻仍以偏粳为主。
Rice (Oryza sativa L.) is an important food crop, which is a constituent of more than 50% of the different varieties of food preparations in China. In order to exploit the utilization of heterosis between rice subspecies, 47 varieties of rice were classified into indica and japonica types firstly, using sequence-related amplified polymorphism (SRAP) and target region amplification polymorphism (TRAP) markers and then simple sequence repeat (SSR) and inter simple sequence repeat (ISSR) markers. Based on differences in the level of polymorphisms detected by the 4 markers-SRAP > ISSR > SSR >TRAP. The average genetic similarity of SRAP is 0.70, TRAP is 0.74, SSR 0.75, ISSR 0.85. which showed highly significant correlation between different markers.The subspecies could be differentiated by cluster analysis while some plants were different in 4 Phylogenetic trees. The result indicated that SRAP and TRAP have potential application in rice research and SRAP has more differentiated ability between varieties that had a near relationship.The javanica rice Lemont and P002 were belong to indica clade. Some wild rice varieties characterized as indica-like or japonica-like in cluster analysis with different markers; however, the cluster analysis of the data sets of the 4 markers combined , all the wild rice varieties were characterized as japonica-like. This indicated that the indica japonica differentiation of Chinese common wild rice varieties were not completed though the japonica-like tendency was happened. )
