为了预测马铃薯蛋白水解物中抗氧化肽的结构,以马铃薯蛋白水解物不同分子量及不同极性组分的氨基酸组成为变量,采用主成分分析对抗氧化肽可能的组成进行预测。并以ABTS自由基清除能力为指标,通过超滤、色谱分离结合质谱分析对马铃薯蛋白水解物中抗氧化肽的结构进行了鉴定。结果表明,主成分分析预测马铃薯蛋白水解物中抗氧化肽序列或片段可能由Tyr、Phe和His三个氨基酸组合而成。这三个氨基酸的组合经验证存在于鉴定得到的马铃薯抗氧化肽Phe-Tyr、Tyr-Tyr、Tyr-Phe-Glu和Pro-Pro-His-Tyr-Phe中。研究表明通过氨基酸分析结合多元统计分析可以为蛋白水解物中抗氧化肽结构的预测提供依据。

o predict the antioxidative peptides present in potato protein hydrolysate, amino acid composition of potato protein hydrolysate fractions with different molecular weight and polarity was used as the variate, and potential peptide sequences were predicted using principal component analysis. Ultrafiltration and chromtograpy seperation was combined with tandem mass spectrum to purify, identify and verify the antioxidative peptides in potato protein hydrolysate based on the ABTS+o scavenging assay. The results showed that the amino acid residues including Tyr, Phe and His were predicted to be the composition in antioxidative peptides. Those three amino acids were present in the sequence of the identified antioxidative peptides Phe-Tyr, Tyr-Tyr, Tyr-Phe-Glu and Pro-Pro-His-Tyr-Phe which were released from patatin. It indicated that combination multiple variate analysis with amino acid composition of protein hydrolysate was able to predict the antioxidative peptide sequence.
