eRF3b-36片段对化学诱导的小鼠肝癌的抑制作用研究

目的 研究eRF3b-36片段对化学诱导的小鼠肝癌的抑制作用及其作用机制。方法 通过DEN+CCL4+乙醇联合诱导建立小鼠肝癌模型,观察不同剂量eRF3b-36片段对肝脏肿瘤发生发展、肝组织病理学改变、血清肝功能指标、肝癌标志物及细胞增殖相关因子mRNA表达水平的影响。结果 eRF3b-36片段可降低肝脏肿瘤数目、改善肝组织病理学改变、降低血清肝功能指标ALT及AST的水平、降低肝癌标志物AFP、GPC3 mRNA的表达以及细胞增殖相关因子Ki-67、Myc、Cyclin D1、Cyclin E1 mRNA的表达。结论 eRF3b-36片段对化学诱导的小鼠肝癌具有抑制作用,其机制可能与抑制细胞增殖有关。
Objective To study the inhibitory effect of eRF3b-36 fragment on chemically induced hepatocarcinoma in mice and its mechanism. Methods The mice hepatocarcinoma model was induced by DEN+CCL4+ethanol, the effects of different doses of eRF3b-36 fragment on the development of liver tumors, the pathological changes of liver tissues, serum liver function indexes, the relative mRNA expression levels of hepatocarcinoma biomarkers and cell proliferation related factor were observed. Results The eRF3b-36 fragment reduced the number of liver tumors, improved the pathological changes of liver tissues, and reduced the serum ALT and AST levels, the relative mRNA expression levels of hepatocarcinoma biomarkers AFP, GPC3 and cell proliferation related factors Ki-67, Myc, Cyclin D1, Cyclin E1. Conclusion eRF3b-36 fragment can inhibit chemically induced hepatocarcinoma in mice and its mechanism may be related to the inhibition of cell proliferation.
