Determination of Huperzine A in Huperzia serrata Extract by High Performance Liquid Chromatography

建立了一种高效液相色谱测定千层塔提取物中石杉碱甲含量的新方法,首次将L-氨基酸引入生物碱高效液相色谱检测流动相。色谱柱:Agilent Zorbax SB-C18(4.6mm×150mm);流动相:甲醇-含1mmol/L L-赖氨酸的水溶液(50:50,v/v);检测波长:310nm;流速:0.6ml/min。结果表明在2~20μg /ml范围内石杉碱甲的量与其峰面积呈良好的线性关系;平均加样回收率为99.8%。该方法灵敏、准确、选择性好。

rapid and selective HPLC method for determination of Huperzine A in Huperzia serrata Extract has been developed and validated. Huperzine A was dissolved 0.01mol/L HCl, chromatographed on a Agilent Zorbax SB-C18(150mm×4.6mm i.d.,5µm) column,with a mobile phase consisting of methanol-1 mmol/L L-Lysine water solution(50:50,v/v),and detected at 310 nm.The UV peak areas were used for quantitation of Huperzine A content.The coorelation coefficient (R2) of the calibration was 0.999 over the range of 2~20 ug/ml.The method was successfully applied to characterize and determinate the Huperzine A in Huperzia serrata Extract.
