GIRK1基因在宫颈癌中的表达及临床意义
Expression of GIRK1 mRNA in cervical cancer
目的 探讨内向整流型钾离子通道(GIRK1)在子宫颈癌组织中的表达及临床意义。方法 采用逆转录-聚合酶链反应(RT-PCR)方法检测60例宫颈癌组织、10对经放化疗前后的宫颈癌组织、10例正常宫颈组织中的GIRK1mRNA的表达情况。结果 GIRK1mRNA在正常宫颈组织中表达的灰度值比值的平均值0.0252与宫颈癌组织中的1.5472相比,有显著性差异(P<0.01)。GIRK1mRNA在高分化癌中表达为0. 9855、中分化为0.8564,与低分化的1.8627相比,有显著性差异(P<0.05)。GIRK1mRNA在宫颈癌Ⅰ-Ⅱ期中表达为 0.9896,在宫颈癌Ⅲ-Ⅳ期表达为1.9742,两者有显著性差异(P<0.01),GIRK1mRNA在放化疗前的宫颈癌组织中表达为l.3710与放化疗后的0.2939相比,有显著性差异 (P<0.01)。结论 GIRK1基因参与宫颈癌细胞恶性增殖过程,GIRK1基因与宫颈癌的恶性进展有关、与宫颈癌的临床分期及病理分级呈正相关,宫颈癌放化疗可降低GIRK1基因的表达。
Objectives To study the expression of GIRK1 mRNA in cervical cancer. Methods Reverse transcription polymerase chain reaction(RT-PCR) was used to detect the expression of GIRK1 mRNA in 60 patients with cervical cancer,10 patients with cervical cancer before and after radiotherapy and chemotherapy, and 10 patients with normal cervical tissue. Results the ratio of gray level of GIRK1 mRNA was 0.0252 in normal cervical tissue,1.5472 in cervical cancer,there were significant differences between them (P<0.01).The ratio of gray level of GIRK1 mRNA was 1.8627 in the poor differentiation cancer, higher than that of the middle and high differentiation cancer 0.9855 and 0.8564(P<0.05). the ratio of gray level of GIRK1 mRNA was 0.9896 in stages Ⅰ~Ⅱ,1.9742 in stages Ⅲ~Ⅳ,there were significant differences between them (P<0.01).GIRK1 mRNA was 1.3710 before radiotherapy and chemotherapy, but 0.2939 after radiotherapy and chemotherapy, they have significant differences(P<0.01). Conclusion GIRK1 mRNA participates in the proliferation of cervical cancer cell, and plays an important role in tumorgenesis of cervical cancer. GIRK1 mRNA have a positive correlation with clinical stages and pathological grades of cervical cancer. The expression of GIRK1 mRNA in cervical cancer was decreased after radiotherapy and chemotherapy.
