JNK在慢性移植物抗宿主病狼疮样小鼠肾组织的表达研究

Objective: To explore expression and significance of c-Jun N-terminal kinase （JNK)in renal tissue of murine with chronic graft-versus-host disease (cGVHD) lupus nephritis. Methods: Mice models of cGVHD lupus nephritis were establised according to relative literatures. Mice were randomly divided into four groups，control group of twelth week (A，n=6)，conctol group of sixteenth week (B，n=6)，model group of twelfth week (C，n=6) and model group of sixteenth week (D，n=6).The mice of group A and C were killed twelve weeks later and the mice of group B and D were killed sixteen weeks later respectively. Histological changes of kidney were observed by HE and Masson staining. The protein and mRNA expressions of JNK and p-JNK in renal tissue were detected by immunohistochemistry, Western blot and RT-PCR respectively. Proteinuria was evaluated by biuret chromometry. Results: Compared with the control groups，the urinary protein excretion，protein and mRNA expression of JNK and p-JNK were increased significantly in the 12w model group respectively (P<0.01) and they were increased more significantly in the 16w model group (P<0.01). Conclusion: The expression of JNK in renal tissue of murine with cGVHD was increased significantly，JNK maybe take an important part in the pathogenesis of murine lupus nephritis.
