反复热性惊厥过程中CBS/H2S系统对HO-1/CO系统的作用

目的 研究反复热性惊厥(febrile seizures,FS)过程中,内源性CBS/H2S系统对HO-1/CO系统的作用。方法 21日龄 SD大鼠,采用热水浴诱导大鼠惊厥,隔日 1 次,共 10 次。大鼠随机分为四组: 正常对照组、高热惊厥组、高热惊厥, FS + NaHS(硫化氢外源性供体)组,FS + HA(磷酸吡多醛-5`-磷酸-依赖性酶包括胱硫醚-β-合成酶抑制剂)组。采用分光光度计法测定大鼠血浆中CO含量;用原位杂交、免疫组化检测方法对海马HO-1 mRNA及蛋白表达进行定位及半定量分析。结果 硫化氢供体的干预可以增强HO-1/CO系统的表达,而磷酸吡多醛-5`-磷酸-依赖性酶包括胱硫醚-β-合成酶抑制剂可以抑制HO-1/CO系统的表达。结论 在反复热性惊厥过程中,气体信号分子H2S对HO-1/CO系统的表达具有重要的调节作用。

he aim of this study is to explore the effect of hydrogen sulfde on heme oxygenase-1/ carbon monoxide (HO-1/CO) system in recurrent febrile seizures (FS) in rats. On a rat model of recurrent FS, the measurement of plasma CO, the expressions of HO-1 were examined after treatment with NaHS, a donor of hydrogen sul?de, or hydroxylamine, an inhibitor of cystathionine b-synthase. The results found that sodium sulfhydrate (NaHS, a donor of hydrogen sul?de) increased the level of plasma CO, up-regulated the expression of HO-1. Hydroxylamine (an inhibitor of cystathionine b-synthase) decreased he level of plasma CO, down-regulated the expression of HO-1. These results suggest that H2S plays a regulatory role through modulating HO-1/CO system in the pathogenesis of recurrent FS.
