清肺化痰、益气活血对肺纤维化大鼠不同时点TGF-β1mRNA表达的影响

目的 比较清热化痰、益气活血、两法联合、两法分阶段四组中药对肺纤维化大鼠不同时间、不同吞噬细胞TGF-β1mRNA表达的影响。方法 将77只雄性SPF级Wistar大鼠随机分为7组,即假手术组、模型组、清肺化痰组、益气活血组、两法联合运用组、两法分阶段运用组及氢化可的松组,建立AM、IM细胞模型。用原位杂交方法检测TGF-β1mRNA表达水平,积分光密度法计算巨噬细胞TGF-β1mRNA阳性面积。结果 7天时AM和28天时IM的 TGF-β1mRNA阳性细胞率显示,中药各治疗组与模型组有显著性差异(P<0.05)。结论 清肺化痰、益气活血两法均可通过减少AM、IM分泌TGF-β1,抑制肺泡炎和纤维化的进程。

Objective To study the effect of QFHT Principle,YQHX Principle, two method associated usages and two method to induct by stages on the model which idiopathic pulmonary fibrosis rat was formed by bleomycin injection via trachea.And explore the academic gist and functional mechanism of preventing the TGF-β1mRNA expression excreted by Alveolar Macrophage and Intercrop Macrophage.Methods Seventy-seven male SPF rate Wistar rats were randomly divided into 7 groups,11 in each group,i.e.the normal control group,the model group,the QFHT group,the YQHX group,the two method associated group,the two method to induct by stages group and the hydrocortisone group. Establishment of the cell model of Alveolar Macrophage and Intercrop Macrophage, and the cultured cells were stained with nucleic acid hybridization in situ to observe the TGF-β1mRNA expression.Results The impacts on TCF-β1mRNA: the proportion of TCF-β1mRNA positve cells on the 7th day AM and the 28th day IM show that TCM therapy can evidently inhibit the alveolar macrophage and lung cell of Leydig to expressed through TCF-β1mRNA,therefore, the airsacculitis and fibrotic process are inhibited(P<0.05).Conclusion The mechanism of QFHT Principle,YQHX Principle, two method associated usages and two method to induct by stages on preventing pulmonary fibrosis is to inhibit the process of airsacculitis and pulmonary fibrosis. And the performance of the mechanism is showed by decreased the expression of TGF-β1mRNA in alveolar macrophage and alveolar interstitial macrophage.
