畜禽的产肉潜力与其肌纤维的数量密切相关。鸟类与哺乳类骨骼肌纤维的数量均取决于胚胎期成肌细胞增殖分化的程度,因此研究胚胎期成肌细胞增殖分化及其调控机制不仅有重要的学术价值,而且对提高畜禽的产肉潜力具有重要的意义。为探讨胚蛋注射CDCA对禽类骨骼肌发育的影响,本研究选用已孵化3d的纯种白洛克鸡胚蛋240枚,随机分成四组,分别注射PBS(空白组)、DMSO(阴性对照组)、500ng/50μL CDCA(低剂量组)、1000ng/50μL CDCA (高剂量组)。结果表明,胚蛋注射低剂量CDCA显著提高了D42时肉鸡体重、腿肌重、肝脏重、E16和DOH(岀壳当天)时单位面积腿肌肌纤维数目;注射高剂量CDCA显著提高了DOH时雏鸡体重和D42时肉鸡肌纤维直径;RT-PCR结果显示,DOH和D42时500ng/50μL CDCA组腿肌中decorin和成肌相关基因(Myf-5、MyoD、Myogenin)mRNA表达水平显著上调,表明胚蛋注射CDCA显著促进禽类胚胎期骨骼肌纤维的发育,提高产肉潜力。

he meat production potential of animal is closely related to muscle fiber number. The number of muscle fibers in avian and mammal skeletal muscle was determined by the degree of embryonic myoblast proliferation and differentiation, therefore the research on skeletal muscle development in avian embryos not only has important academic value,but also has great significance in improving the meat production potential.In order to explore the effect of in ovo infection of CDCA on avian skeletal muscle development, 240 White Plymouth Rocks breeder eggs on day 3 of embryonic age were selected and allocated to four groups randomly, which were injected in albumen with 500ng/50μL CDCA (low- dose group), 1000ng/50μL CDCA (high-dose group), DMSO (negative group) or 50μLPBS (control group). The results showed in ovo injection with 500 ng of CDCA significantly increased body weight, liver weight, leg muscle weight of broiler on day 42 and leg muscle fiber density on E16 chick embryo and DOH (day of hatch). There was an increase in chick weight on DOH and leg muscle fiber diameter of broiler on day 42 in the group of injected with 1000ng/50μL CDCA. Results of RT-PCR showed that the mRNA expression levels of decorin and myogenic regulatory genes (Myf-5, MyoD and Myogenin) were significantly upregulated in leg muscle of low-dose group on DOH and D42. These results suggested in ovo injection of CDCA could promote development of muscle fiber and improve meat production in broilers.
