目的 研究隐丹参酮对人宫颈癌细胞株Hela的放射增敏作用及对细胞周期和凋亡的影响。方法 MTT比色法检测不同照射剂量0、1、2、4、8Gy下,隐丹参酮(4.03µg/ml)对Hela细胞的增殖抑制作用;克隆形成实验分析,多靶单击拟合模型方程计算放射增敏比,评价增敏效果。流式细胞仪检测隐丹参酮对Hela细胞周期和凋亡的影响。结果 给药隐丹参酮对Hela细胞毒性有明显照射剂量依赖性。隐丹参酮抑制细胞20%的浓度(4.03µg/ml)对Hela细胞具有放射增敏作用,克隆形成法得到其增敏比为1.4。隐丹参酮可使Hela细胞周期的构成发生明显的变化并诱导细胞凋亡,使S期的细胞比例降低,出现明显的Go/G 1期阻滞。结论 隐丹参酮对宫颈癌细胞系Hela有较好的细胞毒性和放射增敏作用,其增敏机制可能是改变细胞周期构成并诱导细胞凋亡,使Hela细胞生长停滞在G0/G1期,使S期细胞比例降低,使肿瘤细胞的增殖速度减慢,从而加强射线对肿瘤细胞的杀伤能力。

Objective To investigate the radiosensitization effects, the cell cycle and apoptosis induction of cryptotanshinone on Hela cell line of cervical cancer. Methods The MTT assay was used to detect the growth inhibition rates of Hela cells at different doses of 0,1,2,4,8Gy which cultured with cryptotanshinone (4.03µg/ml). With multi-target single-hit model, The radi-osensitization effect was investigated by clone formation assay through sensitizing enhancement ratio(SER). The cell cycle distribution and apoptosis were tested by flow cytometry. Results At different irradiation doses,the cryptotanshinone had cytotoxicity on Hela cells, which were clearly dose-dependent. The IC20 of cryptotanshinone(4.03µg/ml) indicated the radiosensitization on Hela cell line. Further clone formation assay indicated that the sensitization enhancement ratio was 1.4.Cell treated with cryptotanshinone, which indicated significant Go/G 1 phase arrest and a decrease in S phase. Conclusions Cryptotanshinone had significant cytotoic and radio- sensitization effects on Hela cell. One of the mechanism maybe that it might have significant effects on cell cycle and induce apoptosis of Hela cell, have significant on G0/G1 phase arrest and a decrease in S phase which cells were more sensitive to radiation.
