马铃薯 Y 病毒(Potato virus Y, PVY)是马铃薯 Y 病毒科(Potyviridae)、马铃薯 Y 病毒属(Potyvirus)的代表种,是生产上严重危害烟草等茄科作物产量和品质的主要植物病毒之一。本研究使用生物信息学方法在全基因组水平上对烟草半胱氨酸蛋白酶家族以及靶向该基因家族的 miRNAs 进行鉴定,分析其在 PVY 感染后的表达特性。结果表面,栽培烟草中共鉴定到 70 个半胱氨酸蛋白基因家族成员,根据其系统发育特征和结构进行分类可分为 5 个亚家族(C1A、C2、C12、C13、C14)。基于 Small RNA 测序,发现 28 个半胱氨酸蛋白酶基因受 51 个 miRNAs 调控,其中 7 个在 PVY 感染后与 miRNAs 负相关。qRT-PCR 结果证明 PVY 感染后,有 15 个半胱氨酸蛋白酶基因表达量显著上调。研究结果有助于深入理解烟草半胱氨酸蛋白酶基因家族功能及其与相应miRNAs的互作,以及通过互作调控烟草对 PVY 的响应。

Potato virus Y (PVY) is a representative species of Potyviridae and Potyvirus, and is one of the major plant viruses that seriously harm the yield and quality of tobacco and other solanIdentification of the cysteine protease family and correspondingaceous crops in production. In this study, the cysteine protease gene family of Nicotiana tabacum L. and their corresponding miRNAs were identified at the genome-wide level. And then analyze their expression characteristics after PVY infection. The results showed that a total of 70 cysteine protease genes were identified in Nicotiana tabacum L. genome, which could be divided into five subfamilies (C1A, C2, C12, C13 and C14).Based on the sequencing results of small RNA-seq, 51 miRNAs were found to be targeted to 28 members of cysteine protease gene family, 7 of the cysteine protease genes were negatively correlated with miRNAs after PVY infection. The results of qRT-PCR showed that 15 cysteine protease genes were significantly up-regulated after PVY infection. This study will be helpful for better understanding the function of cysteine protease gene family in Nicotiana tabacum L. and the interaction of the family genes with their corresponding miRNAs, and how this interaction regulates the response of Nicotiana tabacum L. to PVY.
