目的:探讨不同浓度的釉基质蛋白(enamel matrix proteins, EMPs)对人牙周膜细胞增殖及牙骨质相关蛋白基因表达的影响。 方法:分别用浓度为0(对照组),50, 100, 200, 300 μg/ml的EMPs作用于培养的人牙周膜细胞(periodontal ligament cells, PDLCs)。在培养的第1、3、5、7天用MTT法检测细胞增殖活性;在培养的第7天,用荧光实时定量RT-PCR法检测各组细胞牙骨质附着蛋白(cementum attachment protein, CAP)和牙骨质蛋白-23(cementum protein, CP-23)的mRNA表达量。结果:EMPs浓度在50-200 μg/ml范围内促进人PDLCs增殖,然而在300 μg/ml时抑制细胞增殖;EMPs浓度在100-200 μg/ml范围内可以明显增加CAP和CP-23的mRNA表达量,且以200 μg/ml 的促表达效果最佳。 结论:EMPs在一定浓度范围内能够促进人PDLCs增殖以及CAP和CP-23表达,提示EMPs可能通过促进细胞增殖和牙骨质相关蛋白的表达,从而促进牙骨质的形成。

Objective: To observe the effects of enamel matrix proteins (EMPs) on proliferation and gene expression of cementum related protein in human periodontal ligament cells. Methods: Human periodontal ligament cells were treated with EMPs at concentrations of 0 (control),50, 100, 200, 300μg/ml. At day1, 3, 5, 7, MTT assay was used to measure cell proliferation; At day 7, real-time RT-PCR was used to detect mRNA expression of CAP and CP-23. Results: EMPs significantly increased cell proliferation at concentrations of 50-200 μg/ml, while it inhibited cell proliferation at concentration of 300 μg/ml. The mRNA expression of both CAP and CP-23 at EMPs concentrations of 100-200 μg/ml were higher than that of the control, and the best concentration was 200μg/ml. Conclusion: EMPs can promote cell proliferation and increase the gene expression of CAP and CP-23. It suggests that EMPS maybe enhance cementum regeneration through promoting hPDLCs' proliferation and cementum related protein production.
