目的 观察金黄色葡萄球菌α-毒素(α-Toxin)对Balb/c小鼠肺微血管内皮细胞(LMEC)水通道蛋白-1(AQP1)表达的影响。方法 将实验组小鼠15只腹腔注入α-Toxin后根据6h、10h、24h取材时间随机将它们分为α-Toxin 6h组、α-Toxin 10h组和α-Toxin 24h组。取左肺行病理HE染色和免疫组化染色,检测肺组织病理变化情况和AQP1表达的变化。结果 α-Toxin各组肺泡间隔增宽、水肿,炎细胞浸润,肺泡结构破坏,以α-Toxin 10h组最为明显。免疫组化α-Toxin各组AQP1表达下降(p<0.05)。其中以α-Toxin 10h组最为明显。α-Toxin 6h组和α-Toxin 10h组及α-Toxin 10h组和α-Toxin 24h组均有显著性差异(p<0.05)。结论 AQP1表达于LMEC。金葡菌α-毒素可致小鼠肺LMEC的AQP1表达下降,随着炎症的减轻,AQP1也随之表达增加,进而恢复正常。

Objective: To observe the effect of S.aureus alpha toxin on pulmonary aquaporin-1 (AQP1) expression in mice. Methods: Balb/c mice weighted 18~27g was divided into alpha toxin-6h group, alpha toxin-10h group and alpha toxin-24h group according to the time point after injecting alpha toxin peritoneally. Immunohistochemistry stain for pulmonary AQP1 was done after execution. Results: Alveolar wall showed edema, inflammatory cell infiltration in all alpha toxin groups, especially in alpha toxin-10h group. AQP1 expression decreased in these groups, in which alpha toxin-10h group was more obvious. Conclusion: AQP1 is mainly localized on lung microvascular endothelial cell. AQP1 expression decreased after mice exposed to S.aureus alpha toxin, while down-regulated AQP1 returned to normal level overtime.
