Method for evaluating activity of neutralization antibodies against SARS-CoV-2
新型冠状病毒SARS-CoV-2引起的全球疫情尚未完全结束。通过疫苗接种建立有效免疫屏障仍是个体防护的关键措施,而血清中中和抗体活性是个体免疫保护力的一项重要指标。本文基于SARS-CoV-2病毒S蛋白与宿主细胞ACE2受体的作用机制,开发了一种体外快速评估SARS-CoV-2抗体中和活性的新方法。在该方法中,我们将S蛋白的受体结合基序(Receptor Binding Motif,RBM)与一段DNA扩增报告序列共价偶联,形成了用于荧光定量PCR检测的重组探针RBM-DNA。同时,我们构建了五聚体RBM结合蛋白2B-LCB1,并将其与固相磁珠偶联获得RBM-DNA探针的快速捕获元件Mag-LCB1。最后,利用qPCR方法实现对RBM-DNA探针与Mag-LCB1相互作用的定量检测,并应用于SARS-CoV-2中和抗体活性的评估。实验结果表明,该方法可以有效检测疫苗接种人群血清中和抗体活性,其对中和抗体的有效检测限LOD为4.05 μg/mL。本文提供了一种全新的SARS-CoV-2中和抗体活性检测方法,其对后疫情时代个体免疫接种保护力评估具有重要意义。
he pandemic caused by the novel coronavirus SARS-CoV-2 has not completely ended. Establishing effective immunity against the virus through vaccination is still the key measure for individual protection, and the activity of neutralizing antibodies in circulation is an important marker of the vaccine-induced immunity in individuals. In this work, we developed a novel method for rapid evaluation of neutralizing activity of SARS-CoV-2 antibodies, based on the specific interactions between the spike (S) protein of the virus and the ACE2 receptor of host cells. In this method, the receptor binding motif (RBM) of the S protein was covalently conjugated with a reporter DNA to form a hybrid amplification probe (RBM-DNA) for quantitative PCR analysis, while a pentameric RBM-binding protein (2B-LCB1) was constructed, which was further immobilized on the magnetic beads as a rapid capturer (Mag-LCB1) of RBM-DNA probes. Finally, the interaction between RBM-DNA and Mag-LCB1 was quantatively determiend by qPCR analysis, based on which the activty of neutralization antibodies can be indirectly quantified. The assay validation data showed that this assay was able to estimate the activity of serum neutralization antibodies from vaccinated individuals, and the limit of detection (LOD) for the neutralizing antibody was 4.05 μg/mL. In summary, this work provided a novel method for detecting the immune activities of SARS-CoV-2 neutralizing antibodies, and we envisage that it will prove to be an useful tool for evaluating vaccine-induced immunity of individuals in the post-pandemic era.
