目的 研究TRAIL诱导骨肉瘤细胞株HOS-8603细胞凋亡的作用以及此过程中c-myc基因表达规律,探讨TRAIL诱导骨肉瘤细胞凋亡的可能机制。方法 传代细胞分为对照组、TRAIL组和TRAIL+caspase3抑制剂组。分别采用透射电镜(TEM)观察细胞凋亡的形态;MTT法检测细胞的抑制率;流式细胞仪(FCM)分析细胞凋亡比例;RT-PCR检测TRAIL受体(receptor,R)及c-myc基因的表达水平。结果 对照组、TRAIL浓度分别为0.1、0.5、1、2μg/ml的TRAIL组及TRAIL+caspase3抑制剂组(0.5μg/mlTRAIL+100μM Z-DEVD-FMK)凋亡细胞比例分别为2.53%、7.33%、13.18%、24.19%、53.75%、7.67%;RT-PCR c-myc条带光密度和β-action的比值分别为0.98、0.82、0.67、0.59、0.49、0.88。RT-PCR检测到TRAIL R1~3的表达。TEM观察到细胞核固缩,染色质浓缩并边缘化为新月形胞核。结论 在体外实验中,TRAIL可通过诱导细胞凋亡而产生抗骨肉瘤的效应,这种效应是caspase依赖性的。在此过程中,c-myc和caspase3存在着反馈抑制调节的可能性。c-myc可能为TRAIL诱导HOS-8603凋亡调控机制中的一个关键环节。

objective Through investigating the effect of TRAIL induced apoptosis of osteosarcoma cell line HOS-8603 and the expression of c-myc during the course,to explore the potential mechanism of this effect.Methods HOS-8603 cells were cultured ,then divided into three groups :control group,TRAIL groups and TRAIL+caspase3 inhibitor group.The morphologic character of apoptosis was observed by TEM.The inhibitive rate was analysed by MTT assay and the apoptosis rate was done by FCM.The expression of TRAIL receptor and c-myc was examined by RT-PCR.Results Apoptosis rate of control group, TRAIL groups(including 0.1
