目的:探讨HBx蛋白对HepG2细胞增殖的影响及其对Wnt/β-catenin信号通路的影响。方法 :利用已成功构建的高表达HBV X 基因的HepG2-HBx细胞株作为实验组,以HepG2-mock细胞株和HepG2细胞株作为对照组。克隆形成实验检测各组细胞增殖能力;以β-actin为内参 ,半定量RT- PCR法检测增殖相关基因β-catenin、CyclinD1及C-myc mRNA等表达量变化。采用SPSS15软件进行统计学分析。结果 : 克隆形成实验显示,与HepG2-mock细胞株和HepG2细胞株相比,稳定表达HBx的HepG2/HBx细胞的细胞增殖能力明显加快(P<0.05)。RT-PCR结果显示HepG2-HBx细胞中β-catenin、CyclinD1及C-myc mRNA相对表达量较HepG2-mock细胞株和HepG2细胞株显著增高 (P <0 . 0 5 )。结论 : HBx因可能通过激活Wnt/β-catenin信号通路促进HepG2细胞的增殖。

Objective:To investigate the effects of Hepatitis B virus X protein on cell proliferation and Wnt/β-catenin signaling pathway of HepG2.Methods: Human hepatocellular carcinoma cell lines of HepG2-HBx ,which can stably express HBx gene were used as the experiment group,paralleled with the HepG2-mock and HepG2. The effect of HBX stably expressing on the proliferation of HepG2 cells was examined by colony formation assay.The expression levels of β-catenin,CyclinD1 and C-myc mRNA were measured by semi-quantitative RT-PCR in HepG2/HBx and control group,respectively. Results: Compared with the control group, the proliferation of HepG/HBx increased significantly.The expressions of β-catenin,CyclinD1 and C-myc mRNA were significant higher than control group(P<0.05). Conclusion:Over-expression of HBx can promote the proliferation of HepG2 cells via the activation of Wnt/β-catenin signal pathway. ?
