Study on the vitro release of GM-CSF Microspheres

运用多糖"水相-水相"乳液法将GM-CSF在温和的条件下包封于耐受苛刻条件的多糖微粒中,然后将多糖颗粒通过亲水"油相"为连续相的无水乳化法包封于PLGA 微球中。扫描电镜图像显示微球的粒径在10 μm左右,分布较为均匀,GM-CSF包封率为83.79%。体外释放试验表明该微球突释小,能够平稳释放一个月,31天的累计释放率达到了95%。

In this paper, a polysaccharose aqueous phase-aqueous phase emulsion was used to encapsulate GM-CSF into polysaccharide particles which are able to withstand the harsh conditions. And then these glassy particles were microencapsulated into PLGA microspheres using solid-in-oil-in-hydrophilic oil(S/O/hO). The results showed that size of the microspheres was about 10 μm in diameter and had a narrow size distribution.Meanwhile,the release test in vitro provided evidence that the microspheres, with an encapsulation efficiency of 83.79 % and low burst release, achieved a sustained release of about one month.
