慢性砷染毒对人支气管上皮细胞中组蛋白H3第27位赖氨酸三甲基化的影响

目的:研究慢性亚砷酸钠染毒人支气管上皮细胞(BEAS-2B)对组蛋白H3第27位赖氨酸(H3K27)三甲基化的影响。 方法:用不同浓度的亚砷酸钠(0、0.1、0.5和1.0 μM)染毒BEAS-2B细胞,分别在染毒时间为4、8、12和16周时提取组蛋白,通过Western blot检测细胞中H3K27三甲基化的水平。 结果:与对照相比,在0.1和0.5 μM的亚砷酸钠染毒4和8周后H3K27三甲基化下降,而在1.0 μM的亚砷酸钠染毒4和8 周后无明显差异,在0.1、0.5和1.0 μM的亚砷酸钠染毒12周后H3K27三甲基化均下降,而在染毒16周后无明显变化。 结论:慢性砷染毒可诱导与基因沉默有关的H3K27三甲基化下降。
Objective: The objective of the study was to investigate the effect of chronic exposure to arsenite on global histone H3 lysine 27 (H3K27) tri-methylation in normal human bronchial epithelial BEAS-2B cells. Methods: BEAS-2B cells were treated with arsenite at 0, 0.1, 0.5, and 1.0 μM for 16 weeks. At 4, 8, 12, and 16 weeks after exposure to arsenite, histones were extracted and H3K27 tri-methylation was examined by Western blot. Results: Compared with untreated cells, H3K27 tri-methylation was decreased after exposure to 0.1 and 0.5 μM arsenite for 4 and 8 weeks, but no significant change was observed in the cells treated with 1.0 μM arsenite for 4 and 8 weeks. At 12 weeks, H3K27 tri-methylation was decreased by 0.1, 0.5, and 1.0 μM arsenite, but there was no significant change at 16 weeks. Conclusion: Chronic exposure to arsenite decreased global H3K27 tri-methylation in BEAS-2B cells.
