猪SOCS1基因多态性分析
Polymorphism analysis of porcine SOCS1 gene
本研究选择SOCS1作为猪抗病育种候选基因,利用直接测序法寻找SOCS1基因多态性,并在民猪和长白猪群体中进行基因频率和多态信息含量分析。研究发现在SOCS1基因5'UTR区域存在一个C/T突变,建立了针对该突变的DpnI PCR-RFLP检测方法。该位点在民猪和大白猪群体中均存在3种基因型,C等位基因频率分别为0.42和0.51。多态信息含量分析发现,该位点在民猪和长白猪群体中均为中度多态。本研究为下一步进行多态位点和抗病性状的关联分析奠定基础。
In this study, SOCS1 gene was selected as a candidate gene for porcine anti-disease traits. Direct sequence method was used to detect the polymorphism of SOCS1 gene. The analysis of gene frequency and polymorphic information content were performed in Minzhu and Landrace pig populations. A C/T mutation in the 5'UTR region of this gene was found and a DpnI PCR-RFLP assay was established to detect this mutation. Statistics analysis showed that there were three genotypes in Minzhu and Landrace populations and the C allele frequency was 0.42 and 0.51 respectively. The polymorphic information content was moderate diversity in both Minzhu and Landrace. Association analysis between this mutation and anti-disease traits in different pig populations will be performed in future research.
