姜黄素对过氧化氢诱导小鼠脑神经瘤细胞氧化损伤的保护作用
Protective effects of curcumin on mouse neuroblastoma Neuro-2A cells against oxidative damage induced by hydrogen peroxide
本文研究了姜黄素对过氧化氢(H2O2)诱导的小鼠脑神经瘤细胞Neuro-2A氧化损伤的保护作用及其机理。将Neuro-2A细胞接种至培养板中,不同浓度姜黄素预孵,继而30 μmol/L H2O2 处理24 h。MTT法测定细胞存活率,核染色法检测细胞凋亡,比色法检测细胞总抗氧化能力,流式细胞术检测线粒体膜电位。结果显示,采用25 μg/mL姜黄素预孵育1 h,Neuro-2A细胞的存活率提高8.5%,细胞凋亡率降低15.1%,总抗氧化能力上升15.9%,罗丹明123 阴性细胞数减少11.81%。由此可见姜黄素对H2O2诱导氧化损伤和凋亡的Neuro-2A细胞具有显著保护作用,其作用机制可能是提高细胞内总抗氧化能力,抑制线粒体膜电位的降低。该发现为氧化应激引起的神经退行性疾病的预防与治疗提供了一条可能的途径。
he neuroprotective effects and its potential mechanisms of curcumin against H2O2-induced oxidative damage in mouse neuroblastoma Neuro-2A cells was studied. Neuro-2A cells were seeded into culture plates, and pretreated with curcumin at different concentrations prior to 30 μmol/L H2O2 exposure for 24 h. Cell viability was measured by MTT assay, cell apoptosis was assessed by nuclear staining, the level of intracellular total antioxidant capacity (TAC) was determined by colorimetric method, and mitochondrial membrane potential was tested by flow cytometry. Results showed that pretreatment with 25 μg/mL curcumin for 1 h increased the percentage of viable cells by 8.5%, reduced the percentage of apoptotic cells by 15.1%, elevated TAC by 15.9%, and decreased the percentage of the Rhodamine 123 negative cells by 11.81%. It is concluded that curcumin is endowed with significant protective effects against H2O2-induced oxidative stress and apoptosis in Neuro-2A cells by its ability to enhance TAC, and inhibit the loss of mitochondrial membrane potential, and might be a therapeutic agent for prevention and treatment of neurodegenerative diseases implicated with oxidative stress.
