血红素加氧酶-1介导Ca2+信号调控的黄瓜不定根发生
Heme oxygenase-1 is involved in Ca2+ signal-regulated cucumber adventitious root formation
应用药理学、生理学及分子生物学技术研究了血红素加氧酶-1(HO-1)在Ca2+信号诱导黄瓜不定根发生中的作用。实验结果表明,HO-1诱导剂氯化血红素和氯化钙(CaCl2)均能以浓度依赖性方式诱导黄瓜不定根发生,且能不同程度的上调HO-1转录本;HO-1的抑制剂ZnPP能够部分抑制CaCl2诱导的不定根发生和黄瓜HO-1基因表达;进一步采用Ca2+螯合剂EGTA及Ca2+通道抑制剂LaCl3和钌红处理,发现其在降低Ca2+ 含量的同时,还能够不同程度的抑制不定根发生及HO-1蛋白表达。上述实验结果表明HO-1可能介导Ca2+调控的黄瓜不定根发生。
In this report, the role of heme oxygenase-1 (HO-1) in Ca2+ signal-induced cucumber adventitious root was investigated by pharmacological, physiological and molecular techniques. Our results showed that both of hemin (an inducer of HO-1) and calcium chloride (CaCl2) could induce cucumber adventitious root formation in a dose-dependent manner. Meanwhile, the up-regulation of cucumber HO-1 gene transcripts was observed. However, ZnPP, an inhibitor of HO-1, was able to inhibit cucumber adventitious root formation and down-regulate cucumber HO-1 gene expression. In a subsequent experiment, we discovered that Ca2+ chelator (EGTA) and Ca2+ channel inhibitors (LaCl3 and RR) treatment not only blocked cucumber adventitious root formation, but also decreased HO-1 protein level. In conclusion, the results presented in this paper suggested the involvement of cucumber HO-1 in the Ca2+-induced adventitious rooting.
