目的:在前期研究肯定Ca2+和肥大细胞在针刺效应产生中的重要作用的基础上,观察络合足三里穴区Ca2+后,穴区肥大细胞的形态学变化,研究钙离子与肥大细胞在针效产生中的作用及其关系,为揭示针效产生的始动机制提供实验依据。方法:复制小鼠急性胃损伤模型,穴位注射不同浓度EGTA络合足三里穴区Ca2+,光学显微镜下观察足三里穴区皮肤、肌肉处肥大细胞数目、脱颗粒的肥大细胞数目及肥大细胞脱颗粒率。结果:(1)胃损伤状态下,针刺可促进穴位处的肥大细胞脱颗粒。(2)络合穴区Ca2+后,针刺不能促进穴区皮肤、肌肉处的肥大细胞脱颗粒。(3)穴位注射不同浓度的EGTA络合穴区Ca2+时,穴位注射EGTA的浓度越高,针刺后穴区皮肤、肌肉处脱颗粒的肥大细胞越少,肥大细胞脱颗粒率越低,反之亦然。结论:在针刺效应产生中,钙离子与肥大细胞具有一定的作用关系。

Objective: To observe the morphological change of mast cells and acupuncture before and after complexing Ca2+ of Zusanli (ST 36), study on the interaction of Ca2+ and mast cells, provide the experimental evidence of initial mechanism of acupuncture effect. Methods: Hydrochloric acid was used to establish the acute gastric injury model in mice. Gastric emptying rate was applied to evaluate the improvement of gastric function by needling ST36. Different concentrations of EGTA were injected into ST36 respectively and the amount of mast cells, degranulated mast cells in skin and muscle tissues of ST36 were counted by microscope . Results: (1) Acupuncture can promote the degranulation of mast cells in gastric injury mice. (2) Degranulation of mast cells can be restrained by complexing the Ca2+ of ST36.(3) The concentration of Ca2+ has the positive relationship with the degranulation rate of mast cells. Conclusion: Ca2+ and mast cells have close relationship and both of them take part in acupuncture effect.
