Expression and significance of p16 in premature rats w ith Chronic Lung Disease Induced by Hyperoxia

目的:探讨p16基因及蛋白在高氧致早产鼠慢性肺疾病(CLD)中表达及意义。方法:建立高氧诱导早产鼠CLD模型, 80只早产鼠被随机分为实验组及对照组,分别应用免疫组化及RT-PCR技术测定肺组织p16 mRNA及蛋白表达。结果: 暴露高氧后的7天,肺组织的p16的基因及蛋白的表达水平开始低于对照组,并且随着高氧暴露时间的延长,p16的基因及蛋白表达水平逐渐减少,于暴露后的21天达最低值。结论: 暴露高氧中早产鼠,其肺组织的p16 mRNA及蛋白的表达水平降低,其低水平的表达状态与高氧致CLD肺纤维化的发生密切相关。

Objective:To explore the expression and significance of p16 in premature rats with chronic lung disease(CLD) induced by hyperoxia. Methods: 80 premature rats were randomly divided into experimental group with CLD induced by hyperoxia, and control group(n=40). The expressions of p16 protein and mRNA were detected by using immunohistochemical methods and RT-PCR. Results:The expressions of p16 protein and mRNA was lower in the experimental group than those in control group at day 7 , and then gradually decreased with extended exposure to hyperoxia ,but reached the lowest levels at day 21 after hyperoxia. Conclusion: The p16 protein and mRNA expressions were downregulated in premature rats exposure to oxygen. Its downregulation was closely correlated with the development of lung fibrosis.
