基于HA和NA基因的共进化网络研究H7N9病毒的致病性
Study of the pathogenicity of H7N9 virus based on the coevolution network of HA and NA genes
HA和NA基因的进化对于H7N9禽流感病毒的致病性有重要作用,已有研究主要集中在HA和NA基因单个位点变异与致病性的关系,关于HA和NA基因共进化对于H7N9禽流感病毒的致病机制的影响还缺乏系统研究。本研究中我们基于HA和NA基因的变异,构建了H7N9流感病毒HA和NA基因的共进化网络;该网络基于连接度可分为高低两簇,其中低连接度的病毒均为高致病性H7N9禽流感病毒,且其特有边对应的核苷酸位点具有很强的特异性;进一步对高/低致病性H7N9禽流感病毒在其12条共有边上所对应的8个共进化位点进行了差异性分析,发现这8个位点所对应的优势氨基酸与致病性显著相关,在不同致病性的H7N9病毒中表现为不同的氨基酸组成;最后,比对目前实验研究所报道的位点,筛选出可能对H7N9病毒的致病性存在影响的三个关键位点:130、143、235(H7编号)。本研究有助于加深我们对H7N9禽流感病毒致病机制的理解,同时也为研究其它亚型禽流感病毒的致病机制提供参考。
Evolution of HA and NA genes plays an important role in the pathogenicity of H7N9 avian influenza virus. Previous studies on the co-evolution of HA and NA genes focused mainly on the relationship between single-site variations and pathogenicity. However, there is a lack of systematic studies on the impact of co-evolution of HA and NA genes on the pathogenicity of H7N9 avian influenza virus. In this study, we constructed a co-evolution network of H7N9 influenza virus HA and NA genes based on their variations. The network can be divided into two clusters based on connectivity, and low connectivity viruses are all highly pathogenic H7N9 avian influenza viruses. The nucleotide sites corresponding to the unique edges of these viruses have strong specificity. Furthermore, we analyzed the differences in eight co-evolutionary sites corresponding to 12 common edges of highly and low pathogenic H7N9 avian influenza viruses, and found that the dominant amino acids at these sites are significantly associated with pathogenicity, and show different amino acid compositions in different pathogenic H7N9 viruses. Finally, we compared with the reported sites in current experimental studies and identified three key sites, 130, 143, and 235 (H7 numbering), that may affect the pathogenicity of H7N9 virus. Our study deepens the understanding of the pathogenic mechanism of H7N9 avian influenza virus and provides reference for the study of the pathogenic mechanism of other subtypes of avian influenza viruses.
