研究仙鹤草活性组分--黄酮组分(FC)和三萜组分(TC)对胰岛素抵抗人肝癌HepG2细胞的糖代谢的调控作用。方法:利用高糖诱导HepG2细胞建立胰岛素抵抗模型,通过2-NBDG荧光探测示踪剂和葡萄糖氧化酶法分别对HepG2细胞葡萄糖的摄取情况和消耗量进行测定,确定稳定的HepG2胰岛素抵抗模型的建立。0.1-2.5μg/mL的FC、TC分别作用胰岛素抵抗细胞24h,分别观察上述活性组分对胰岛素抵抗HepG2细胞葡萄糖摄取情况和葡萄糖消耗的影响;通过RT-PCR法检测细胞胰岛素传导信号通路中胰岛素受体底物IRS-1和JNK基因的表达。结果表明HepG2细胞在55mM浓度的高糖中作用24h,葡萄糖的摄取和消耗量显著减少(p<0.05),实验成功诱导出稳定的人肝癌胰岛素抵抗模型。通过检测FC,TC干预IR模型HepG2细胞后的糖消耗量和摄取情况以及IRS-1和JNK的表达,发现FC,TC组分均有改善胰岛素抵抗的作用。

Studied on the effects of FC and TC of active ingredients from agrimony on glucose metabolism in IR-HepG2 cells . Methods: establish insulin resistance model which HepG2 cells were induced by high glucose.The uptake and consumption of glucose in HepG2 cells were measured by 2-NBDG fluorescence detection tracer and glucose oxidase method respectively to establish the stable HepG2 insulin resistance model. The 0.1-2.5μg / mL FC and TC treated insulin resistance cells for 24 h respectively and observed the effects of active components on the glucose uptake and glucose consumption of insulin resistant HepG2 cells . The expression of IRS-1 and JNK genes in insulin signaling pathway was detected by RT-PCR.The results showed that HepG2 cells had a significant reduction in glucose uptake and consumption (p <0.05) were incubated with 55 mM high glucose for 24 h, and the stable insulin resistance model of human hepatocellular carcinoma was successfully induced. The effects of FC and TC on the glucose consumption and uptake of HepG2 cells and the expression of IRS-1 and JNK were detected, and we found that FC and TC can effectively improve insulin resistance of intracellular.
