绵羊嗜皮菌病PCR诊断方法的建立
Establishment of polymerase chain reaction assay for detection of Dermatophilosis in sheep
摘要
为建立一个PCR检测动物嗜皮菌病的方法。根据Genbank上发表的刚果嗜皮菌属的部分16s-rRNA基因序列,设计了一对特异性引物。经PCR扩增,从发病绵羊皮肤病料提取的基因组DNA得到了大小约500bp的DNA片段。将PCR产物克隆并测序表明,与Genbank上发表的刚果嗜皮菌基因序列的同源性为90.632%。而猪胸膜肺炎放线杆菌、大肠杆菌、葡萄球菌、肠球菌扩增结果为阴性;PCR的敏感性试验显示这对引物能够检测到1ng的DNA,结果表明,此PCR方法特异性好,敏感性高,可用于嗜皮菌病的快速诊断。Abstract
To establish a detection technique for animals dermatophilosis by polymerase chain reaction(PCR)。A pair of primers specific to the 16s-rRNA gene of Dermatophilus Congolensis from Genbank。With the primers,DNA fragments 500bp in length were amplified from the gene of skin focus DNA by PCR。PCR product was extracted and then ligated to PGEM-T-easy vector. After sequencing and comparison homogencity was 90.632%。No amplification was observed from any other bacteria and actinocbacillus tested,including Actinobacillas Pleurpneumoniae ( App)、Escherichia coli(E.coli)、Staphylococcus(Staph)and Enterococcus(Enter). Those showed that the PCR was special. It proved that the PCR could detect 1ng DNA by analysis of the sensitivity of the primers. It was concluded from the above-mentioned results that the established PCR method can be used to diagnose quickly Dermatophilosis。 关键词
