水稻E2F3转录因子在生长发育中的作用

动物E2F 转录因子通过调控DNA 合成和复制相关基因的表达而参与细胞增殖、生长和凋亡过程,特别在调控细胞从G1期进入到S期过程中起着重要作用。水稻E2F生物学功能尚未清楚。本研究从水稻基因组分离、克隆和鉴定了E2F3 基因,研究发现水稻E2F3 的缺失导致植株生长严重受阻,e2f3突变体植株矮小,其根、茎、叶生长明显弱于野生型,且突变体植株生长后期未能开花结实。通过基因回补技术在突变体中表达E2F3 基因则能明显缓解其生长滞后的表型,表明E2F3 在植物生长发育过程中具有重要的调控作用。研究还发现E2F3 的缺失表达可导致TOR 通路相关基因表达量的上调,表明E2F3 参与TOR 通路的调控过程。
he E2F transcript factors are involved in DNA synthesis and replication to regulate the cell cycle in animal cells, especially in G1/S transition. Plant E2Fs also play an important role in cell cycle. Most studies of E2F are focused on Arabidopsis. The knowledge of E2F is still limited in rice. Here we found that rice E2F3 played an important role in plant growth regulation. The loss of E2f3 resulted in the growth arrest showing a smaller size of plant with less leaf number, less tiller numbers and shorter root length than that of wild type plants. The e2f3 plants failed to flowering and seed setting. The growth arrest was rescued by the complementation with E2F3 gene under the control of its native promoter, which demonstrates the role of E2F3 in plant growth. In addition, the loss of E2F3 conferred the elevated expression levels of genes involved in TOR pathway as revealed by quantitative real time PCR. The date suggests that E2F3 may be involved in TOR signaling to regulate plant growth and development.
