IETD连接TRAIL和Smac的双基因溶瘤腺病毒抑制肿瘤增殖的研究

研究利用IETD连接TRAIL和Smac的双基因溶瘤腺病毒对人肿瘤细胞的增殖抑制作用及其诱导凋亡的机制。将TRAIL-(IETD)-Smac表达框插入溶瘤腺病毒载体ZD55,构成靶向性双基因溶瘤腺病毒ZD55-TRAIL-IETD)-Smac。MTT和结晶紫染色实验证明ZD55-TRAIL-(IETD)-Smac对人肝癌细胞株Huh-7、PLC、Bel-7404、SMMC-7721、HepG2、Hep3B, 人结肠癌细胞株SW620,人乳腺癌细胞株Bcap-37,人鼻咽癌细胞株CNE,人宫颈癌细胞株Hela以及人非小细胞肺癌细胞株NCI-H460的增殖有显著的的抑制作用,而对正常细胞QSG-7701 and WI38影响较小;Hoechst33342染核实验证明ZD55-TRAIL-(IETD)-Smac能够诱导多种肿瘤细胞的凋亡,western blot检测了凋亡信号通路关键蛋白的表达;动物实验表明ZD55-TRAIL-(IETD)-Smac对荷NCI-H460肺癌裸鼠肿瘤有显著的抑制作用。因此,上述实验结果显示靶向性双基因溶瘤腺病毒ZD55-TRAIL-(IETD)-Smac具有显著的体内外靶向杀伤肿瘤的能力及对正常细胞无明显毒副作用,为肿瘤的基因病毒治疗提供了参考。
he purpose is to study the growth inhibition effect of tumor cells and the apoptosis mechanism by TRAIL and Smac dual-gene oncolytic adenovirus ZD55-TRAIL-(IETD)-Smac. The expression cassette TRAIL-(IETD)-Smac, in which the TRAIL and Smac genes were linked together by the sequence IETD, was inserted into an oncolytic adenovirus with E1B 55 kDa gene deletion (ZD55) to construct ZD55-TRAIL-(IETD)-Smac. MTT assay and crystal violet experiment suggest ZD55-TRAIL-(IETD)-Smac has an excellent antitumor effect in vitro for human hepatama, cervical, lung, nasopharyngeal and colorectal cancer cell lines but does no or little damage to normal cell lines QSG-7701 and WI38. Tumor cell apoptosis and principal protein of apoptosis pathway induced by the recombinant adenovirus were detected by Hochest33342 staining and western blot analysis, respectively. Furthermore, animal experiment showed that treatment of ZD55-TRAIL-(IETD)-Smac markedly suppressed xenograft tumor growth of nude mice bearing human lung cancer cells NCI-H460. In conclusion, the findings indicated that targeting dual gene-virus ZD55-TRAIL-(IETD)-Smac has the remarkable cancer-killing effects in vivo and in vitro and is a highly promising therapeutic agent for cancer.
