S100A9介导食管鳞癌细胞辐射抵抗的初步研究
Preliminary study on S100A9-mediated radioresistance of esophageal squamous carcinoma cells
目的:探究S100A9在食管鳞癌细胞放射敏感性调控中的作用。方法:通过在人食管鳞癌细胞系TE-1、KYSE-30中过表达或沉默S100A9,构建相应细胞模型;经克隆形成实验确定S100A9对TE-1和KYSE-30细胞放射敏感性的影响;通过流式细胞术分析S100A9对受照射TE-1和KYSE-30细胞凋亡水平的作用;采用Transwell法检测其对食管鳞癌细胞的迁移能力的影响;通过Western Blot和免疫荧光实验检测DNA损伤相关蛋白γ-H2AX表达的变化。结果:S100A9过表达可增加TE-1细胞的克隆形成和迁移,减少细胞凋亡水平和电离辐射后细胞中γ-H2AX蛋白的表达,提示S100A9可促进食管鳞癌细胞的辐射抗性;沉默S100A9可抑制KYSE-30细胞的克隆形成和迁移,增加细胞的凋亡水平、增加电离辐射后细胞中γ-H2AX蛋白的表达,提示靶向抑制S100A9可增强食管鳞癌的放射敏感性。结论:S100A9是食管鳞癌放射敏感性的关键调控因子与潜在的放射增敏新靶点,其作用机制可能与参与调控食管鳞癌细胞中由辐射诱导的凋亡、DNA 损伤等有关,但涉及的具体分子调控机制仍有待进一步深入研究。
Objective: To investigate the effects of S100A9 in regulating the radiosensitivity of esophageal squamous carcinoma (ESCC) cells. Methods: The S100A9 overexpressed or silenced human esophageal squamous carcinoma cell model were constructed using TE-1 and KYSE-30 cell lines, and the clonogenic assay was used to determine the effect of S100A9 on the radiosensitivity of TE-1 and KYSE-30 cells. By applying flow cytometry analysis, the influence of S100A9 in regulating irradiation (IR) -induced apoptosis was assayed in vitro. Transwell assay was applied to detect the effect of S100A9 on migration of ESCC cells. Western Blot and immunofluorescence staining were performed to identify the expression of DNA damage related protein γ-H2AX. Results: Overexpressed S100A9 could increase the clone formation ability and migration of TE-1 cells, while reduce the apoptosis and the γ-H2AX expression after IR exposure, which indicated that S100A9 could promote the radioresistance of ESCC cells. And vice versa. Conclusions: S100A9 might play a crucial role in regulating the radiosensitivity of ESCC, with the mechanism through regulating IR-induced apoptosis and DNA damage of ESCC cells. Nevertheless, the specific molecular mechanisms need further in-detailed investigation.
