中枢肿瘤坏死因子-α对糖尿病大鼠交感神经活动的影响
Brain tumour necrosis factor-α contributes to sympathoexcitation in diabetic rats
本研究旨在探讨中枢肿瘤坏死因子-α(TNF-α)与其合成阻断剂己酮可可碱(PTX)对糖尿病(DM)大鼠交感神经活动的影响。本试验通过将健康雄性SD大鼠随机分为糖尿病干预组(DM+PTX)、糖尿病对照组(DM+VEH)、正常干预组(SHAM+PTX)和正常对照组(SHAM+VEH),并采用高脂高糖饲料喂养4周加链脲佐菌素注射建立糖尿病大鼠模型,干预组通过侧脑室插管给予PTX,对照组给予人工脑脊液(VEH)。4周后,对各组大鼠分别进行体重、血糖测量。应用免疫组化检测大鼠中枢下丘脑室旁核(PVN)内TNF-α表达,并运用电极记录肾交感神经放电情况。所得结果与SHAM组比较,(1) DM组大鼠PVN内TNF-α表达增加,且DM+SHAM组高于DM+PTX组;(2) DM组大鼠肾交感神经放电明显增强;DM+PTX组与DM+SHAM组比较,肾交感神经放电减弱。阻断中枢PVN区TNF-α合成可以明显降低糖尿病时交感神经兴奋性,最终初步证明中枢TNF-α对糖尿病大鼠交感神经活动存在着重要的影响。
he present study was undertaken to investigate the effect of brain TNF-α on the sympathetic nerve activity in diabetic rats. Sprage-Dawley rats (200±30g) were fed with high fat and sugar diet for 4 weeks following by injection of streptozotocin to induce the diabetes mellitus. Rats were implanted with paraventricular nucleus (PVN) cannulae and treated with the cytokine synthesis inhibitor pentoxifylline (PTX, 10 ug/h), or vehicle (VEH) for 4 weeks. Rats were randomly divided into SHAM+VEH, SHAM+PTX, DM+VEH, and DM+PTX groups. Immunohistochemisty were used to detect the expression of TNF-α in the PVN of hypothalamus, renal sympathetic nerve activity (RSNA) was recorded. Compared with sham groups, diabetic rats had higher level of TNF-α in the PVN. ICV treatment with PTX attenuated TNF-α expression in the PVN compared with VEH-treated diabetic rats. Compared with sham groups, diabetic rats had augmented renal sympathetic nerve activity. In contrast, ICV treatment with PTX decreased sympathetic nerve activity. In conclusion, TNF-α in the PVN contributes to sympathoexcitation in diabetes mellitus.
