猪圆环病毒2型(PCV2)是目前危害养猪业的重要病原,可以引起包括仔猪多系统衰竭综合征(PMWS)的多种疾病,目前尚无有效的治疗方法,只能依靠疫苗预防感染。本研究将经过小鼠免疫试验筛选到的表达修饰Cap蛋白的真核质粒pVAX-SynCap(m)转染减毒沙门氏菌SL3263株(SL),构建了重组沙门氏菌SL-pVAX-SynCap(m),与质粒pVAX-SynCap(m)、空质粒pVAX1、SL和PBS进行猪体免疫攻毒保护试验。首免后2周,PCV2抗体阴性,首免后5周pVAX-SynCap(m)和SL-pVAX-SynCap(m)抗体水平分别为1:150和1:280,但均未检测到中和抗体,而其他组PCV2抗体和中和抗体均为阴性。用PCV2 SH株攻毒后,SL-pVAX-SynCap(m)及pVAX-SynCap(m)组仅出现短时间的发热,临床症状较轻,相对日增重明显高于攻毒对照组(P<0.05),其中SL-pVAX-SynCap(m)免疫组在病毒血症、淋巴结病毒载量和病理组织学变化方面明显轻于攻毒对照组。该研究结果表明,减毒沙门氏菌作为载体后提高了质粒pVAX-SynCap(m)对猪体的免疫保护作用,重组沙门氏菌SL-pVAX-SynCap(m)对猪体具有较好免疫保护作用,为研究有效预防PCV2感染的口服疫苗奠定了基础。

he Cap protein was the major immune protein of PCV2, a pathogene which endangered the pig industry in the world. In this study, the recombinant plasmid pVAX-SynCap(m) expressing the codon-optimization Cap protein was transformed into the attenuated S.typhimurium strain SL3263 by electroporation, resulting in a strain SL-pVAX-SynCap(m). To detect the immunogenicity of the plasmids DNA pVAX-SynCap(m) and the recombinant bacteria SL-pVAX-SynCap(m),the piglets were divided into five groups and vaccinated with the plasmids pVAX-SynCap(m) and pVAX,the bacteria SL-pVAX-SynCap(m) and S.typhimurium strain SL3263 and PBS. The results showed that the pigs vaccinated with the recombinant S.typhimurium and recombinant plasmid developed specific anti-PCV2 antibody by ELISA, but no neutralization antibody response. Meanwhile, no humoral responds could be detected in other control groups. After challenged with PCV2-SH, piglets inoculated with pVAX-SynCap(m) and SL-pVAX-SynCap(m) showed mild clinical signs, lower viremia and less gross lesion of lungs, comparing with those in other groups, and that of the SL-pVAX-SynCap(m)-vaccinated group were better than that of the pVAX-SynCap(m)-vaccinated group. These results indicated that SL-pVAX-SynCap(m) could markedly increase the immune responses and provide efficient protection against virulent PCV2 challenge in pigs. The study laid the foundation for the research of PCV2 oral vaccine which can completely prevent pig from PCV2 infection.
