大黄素甲醚影响巨噬细胞[Ca2+]i变化和TNF-α释放的作用特征
EFFECTS OF PHYSCION ON [Ca2+]i AND TNF-α PRODUCTION IN RAT PERITONEAL MACROPHAGES
应用L929细胞系和MTT法检测TNF-α量,同时用细胞[Ca2+]i荧光图像分析系统检测单细胞[Ca2+]i变化动力学,以研究大黄素甲醚(Physcion,Phy)对正常的和细菌脂多糖 (LPS)刺激的大鼠腹腔巨噬细胞(PMφ)释放肿瘤坏死因子(TNFα)和[Ca2+]i动态变化的影响。结果显示,大黄素甲醚对正常PMφ能适度促进其释放TNFα和升高[Ca2+]i,两种效应都随药物浓度增加而增强;同时,大黄素甲醚显著抑制LPS刺激的PMφ过度释放TNFα和升高[Ca2+]i,而两种抑制效应则随着药物浓度的增加而迅速衰减。以上结果说明,大黄素甲醚对PMφ释放TNF-α和升高[Ca2+]i表现出双向调节作用,但其活化正常细胞免疫功能与抑制失控释放炎性细胞因子的剂量依赖关系相反。研究表明,大黄素甲醚调节PMφ的[Ca2+]i变化可能是其调控细胞释放TNFα的信号传导通路中的重要环节。
o explore the role of physcion on increase in [Ca2+]i and TNF-α production in normal and lipopolysaccharide (LPS)-stimulated rat peritoneal macrophages (PMφ), TNF-α production was determined using TNF-α–sensitive L929 cell line and by MTT method. The kinetics of intracellular free Ca2+ concentration ([Ca2+]i) in a single cell were monitored continuously by fluorescence imaging system. The results showed that physcion could elevate TNF-α production moderately, and cause low-amplitude [Ca2+]i increase in normal single PMφ, and the two effects enhance with the increase of the concentration of physcion. Physcion could inhibit both release of TNF-α and increase in [Ca2+]i stimulated by LPS in PMφ, and the two effects low with the increase of the concentration of physcion. These results indicate that the biphase characteristics of modulation of physcion on TNF-α production correlate with those of physcion on [Ca2+]i., but the relations between the effects and the concentration of physcion
