摘要

目的:以LPS诱导的RAW264.7细胞炎症模型,探讨芦荟大黄素的抗炎作用机制。方法:以不同浓度(5μM、10μM、20μM)的芦荟大黄素预处理细胞1h后,给予1μg/mL的LPS诱导24h。以Griss法检测NO的生成量,以Western Blot方法检测iNOS蛋白表达,以qRT-PCR方法检测iNOS mRNA表达。结果:芦荟大黄素个剂量组可显著抑制NO的释放(P<0.01),iNOS蛋白的表达(p<0.01,P<0.001),iNOS mRNA的表达(P<0.05,P<0.01),且均呈现剂量依赖性。结论:芦荟大黄素具有较好的抗炎活性,可抑制一氧化氮的释放,该活性通过抑制一氧化氮合酶的转录表达发挥作用。

Abstract

Objective: To explore the anti-inflammatory mechanism of aloe emodin in LPS-induced RAW264.7 cells. Methods: RAW264.7 cells treated with 1μg/mL of LPS for 24h after pretreated with aloe-emodin for 1h which is three concentrations (5μΜ, 10μΜ, 20μΜ). The amount of NO was detected by Griss assay. Western Blot method to detect the expression of iNOS protein and the expression of iNOS mRNA was detected in qRT-PCR. Results: Aloe-emodin significantly inhibited the release of NO (P <0.01), the expression of iNOS protein (P <0.01, P <0.001), the expression of iNOS mRNA (P <0.05, P <0.01) in a dose-dependent manner. Conclusion: Aloe-emodin has excellent anti-inflammatory activity, and the mechanism depends on inhibit the transcriptional activity of iNOS.
