人肝细胞癌趋化因子受体表达谱

目的:明确人肝癌细胞及组织趋化因子受体表达谱。方法:采用PCR Array技术检测不同人肝癌细胞株及组织中趋化因子受体表达谱;选择具有代表性的趋化因子受体采用免疫组化技术明确其在人肝癌组织中表达及分布情况。结果:7种人肝癌细胞株均可以检测到CCR1、CCR10、CXCR6等趋化因子受体mRNA表达,CCR5、CCR6、CXCR4在部分人肝癌细胞株中有表达;人肝癌组织均可检测到CCR1、CCR5、CCR6、CXCR4、CXCR6等趋化因子受体mRNA表达。免疫组化显示CCR1、CXCR6趋化因子受体表达定位于肝癌细胞,且肝癌组织表达水平高于癌旁肝组织。结论:人肝细胞癌表达多种趋化因子受体,CCR1、CXCR6在体外培养肝癌细胞株及人肝癌组织中均有较高表达。
Objective:To detect expression of chemokine receptors in hepatocellular carcinoma cell lines and tissues. Methods: Gene expression profile of chemokine receptors of human hepatocellular carcinoma cell lines and tissue was determined by PCR Array. The expression and distribution of the typical chemokine receptors was detected by immunohistochemical staining. Results: The hepatoma cells expressed several chemokine receptors. The expression profile of chemokine receptors in different cell lines was varied. CCR1 、CCR10、CXCR6 were detected in all examined hepatoma cell lines, CCR5、CCR6、CXCR4were observed in several cell lines. CCR1、CCR5、CCR6、CXCR4、CXCR6 mRNA could be detected in HCC tissues. Immunohistochemical staining revealed that CCR1、CXCR6 protein were mainly located on the membrane of HCC cells. Conclusion: HCC cells expressed several chemokine receptors.
