肉毒碱脂酰转移酶在高山被孢霉中的沉默
RNA interference of carnitine acyl-transferase in Mortierella alpina
高山被孢霉是一种工业化生产多不饱和脂肪酸的产油真菌。从代谢工程角度看,阻断目标产物的降解途径能够有效提高目的产物积累量。通过分析高山被孢霉脂肪酸氧化途径中相关基因的转录水平,发现肉毒碱脂酰转移酶(CAT)基因与高山被孢霉脂肪酸氧化紧密相关。随后利用RNA干扰技术实现在高山被孢霉中CAT基因的沉默。分析发现,在得到的重组高山被孢霉中, CAT活性下调了33.3%,总脂肪酸积累量达到了细胞干重的38.4%,较野生型有了明显的提高,表明通过抑制β氧化途径可以提高高山被孢霉体内脂质积累量。该研究为推动高山被孢霉在脂质生物合成工业化方面的应用奠定了理论基础。
Mortierella alpina is a type of fungus used for the industrial production of polyunsaturated fatty acids. In metabolic engineering, the inhibition of degradation pathway is an important way to increase the accumulation of the product. In this study, transcription levels of most of the β-oxidation-related genes were analyzed, and carnitine acyl-transferase was speculated to be the key enzyme involved in the oxidation. To suppress the oxidation of fatty acids, RNA interference technology was used to achieve gene silence of the key enzyme in M. alpina. Enzyme activity analysis showed the specific activity decreased by 33.3%. Strains were cultivated for 10 days at 28 C with shaking at 200 rpm. The fatty acid accumulation reached up to 38.4% of DCW in transformants, which is higher than the content in wild-type M. alpina. Our research provided theoretical support for implications for industrial production of fatty acids in M. alpina.
