尿素循环代谢物富马酸调控巨噬细胞炎症机制的研究
Study on the mechanism of urea cycle metabolite fumaric acid regulating macrophage inflammation
尿素循环在巨噬细胞极化过程中具有异质性,不同的极化刺激方式会导致尿素循环具有不同的代谢模式,尿素循环的代谢物富马酸已被证明对巨噬细胞具有重要的调节作用,但机制仍有待确定。本文通过对巨噬细胞诱导极化处理,向培养基中加入通用的可透膜的富马酸类似物富马酸二甲酯,通过ELISA、qPCR检测发现会抑制巨噬细胞促炎因子NO、TNF-a、IL-6以及IL-1b的产生,银染以及IP实验检测发现富马酸二甲酯对iNOS进行琥珀酸化修饰并抑制其活性。综上所述,本文解释了富马酸抑制巨噬细胞促炎因子NO产生的机制,为尿素循环调控巨噬细胞极化提供了新的见解。
he urea cycle is heterogeneous in the process of macrophage polarization, and different polarization stimulation modes will lead to different metabolic patterns of the urea cycle, and fumaric acid, a metabolite of the urea cycle, has been shown to have an important regulatory effect on macrophages, but the mechanism remains to be determined. In this paper, the production of macrophage pro-inflammatory factors NO, TNF-a, IL-6 and IL-1b was inhibited by ELISA and qPCR, and dimethyl fumarate was succinated and inhibited by silver staining and IP experiments. In summary, this article explains the mechanism of fumarate inhibiting the production of macrophage pro-inflammatory factor NO, and provides new insights into the regulation of macrophage polarization by urea cycle.
