雷公藤甲素诱导大肠癌细胞自噬及其信号通路研究
Study of Autophagy and Signaling Pathways of Colorectal Cancer Cells Induced by Triptolide
目的 雷公藤甲素对人大肠癌sw480细胞自噬的影响以及介导自噬的信号通路。方法 体外培养人大肠癌sw480细胞。分别给予不同浓度的雷公藤甲素进行刺激,Western blot 检测自噬相关蛋白LC3B、Atg7和Beclin-1的表达水平。采用Path Scan 检测药物作用后细胞信号通路的变化情况。结果 当雷公藤甲素浓度为40nM时,自噬标志蛋白LC3B、Atg7和Becline-1水平升高(P<0.05)。Path Scan 结果表明,细胞信号通路的成分被EKR1/2, Akt, P53,AMPKα激活。结论 雷公藤甲素可诱导sw480细胞发生自噬,可能是通过影响ERK1/2、Akt、P53和AMPKα通路实现的。
OBJECTIVE To explore the effect of Triptolide on autophagy of colorectal cancer line sw480 and to reveal the signaling pathways. Methods Sw480 cells were cultured and stimulated with Triptolide of different concentrations. Western blot were performed to analyze the level of autophagy-associated proteins-LC3B, Atg7 and Beclin-1. Path Scan Intracellular Signaling Array Kit was used to detect the change of the cell signaling pathways after the stimulation of Triptolide. RESULTS When treated with 40 nM Tripltolide, the levels of autophagy-associated proteins elevated (P<0.05),and the Path Scan showed that EKR1/2, Akt, P53 were up-regulated and AMPKα down-regualted. CONCLUSIONS Triptolide can induce autophagy of sw480 cells,which is likely to be induced by ERK1/2, Akt, and P53 signaling pathways.
