HPLC测定藏药铁棒锤中乌头碱、3-乙酰乌头碱的含量

目的 测定藏药铁棒锤中乌头碱、3-乙酰乌头碱的含量。方法 采用HPLC法，色谱柱为BDS Hypersil C18（250×4.6 mm, 5 μm），流动相为乙腈-0.3 moloL-1碳酸氢铵溶液（56∶44），检测波长231 nm，柱温为30 ℃。结果 乌头碱（r=0.9991）在0.0411~0.3697 mgomL-1，3-乙酰乌头碱（r=0.9996）在0.0040~0.0360 mgomL-1范围内线性关系良好，平均回收率分别为100.65%，101.62%。结论 11批藏药铁棒锤中乌头碱含量在0.072%~0.306%之间，3-乙酰乌头碱含量在0.033%以下。所建立的测定方法操作简便、准确、重复性好，可为铁棒锤药材的质量控制提供参考
Objective To determine the contents of aconitine and 3-acetylacnitine from roots of Tibetan Medicine Aconitum pendulum. Methods A HPLC method was established on a BDS Hypersil C18（250×4.6 mm, 5 μm）with the mobile phase consisting of acetonitrile-0.3 moloL-1 ammonium acid carbonate liquid , the detection wavelength was 251 nm and the column temperature was maintained at 30 ℃. Results The calibration curves showed good linearity in the range of 0.0411~0.3697 mgomL-1 of aconitine, 0.0040~0.0360 mgomL-1 of 3-acetylaconitine,respectively.The average recoveries of aconitine ,3-acetylaconitine were 100.65%,101.62%.Conclusion The content of aconitine is between 0.072%~0.306% and 3-acetylaconitine is under 0.033%.The method is simple, accuracy and repeatability good,can provide a reference for the mass control of Aconitum pendulum.
