急性髓系白血病患者骨髓细胞相关分子标志物表达研究
Study of biomarkers expression related with Bone Marrow Cells in patients with Acute Myelogenous Leukemia
目的:探讨急性髓系白血病患者骨髓细胞相关分子标志物(PTEN、mTOR、NF-kB、CD44、PI3K)表达。方法:选取20例住院患者,抽取骨髓液,先涂片检查骨髓白血病细胞百分比,并分临床缓解(9例)与未缓解(11例)两组。再将剩余的骨髓液以石蜡包埋,常规切片,免疫组化技术检测PTEN、mTOR、NF-kB、CD44、PI3K表达。结果:光学显微镜观察缓解组与未缓解组PTEN、mTOR、NF-kB、CD44、PI3K阳性细胞表达率分别为33.3、33.3、77.8、22.2、0与63.6,18.2,90.9,63.6,0。经统计学处理,PTEN、CD44两组比较,有统计学意义(p<0.05)荧光表达强度与图像分析也得出同样的研究结果。结论:急性白血病骨髓细胞PTEN、CD44高表达可作为急性髓系白血病难治诊断和预后判定的重要参考指标。
Objective:To detect the expression of biomarkers(PTEN、mTOR、NF-kB、CD44、PI3K) related with Bone Marrow Cells in patients with Acute Myelogenous Leukemia.Methods:The immunohistochemical method was used to detect the expression of PTEN、mTOR、NF-kB、CD44、PI3K in 20 patients,which were separated into remission group(9 cases)and no-remission group(11 cases) after calculating the percentage of leukemia cells in bone marrow.Results: According to Optical microscope detection,the positive expression rates of PTEN、mTOR、NF-kB、CD44、PI3K in remission group were 33.3%,33.3%,77.8%,22.2%,0.Meanwhile,in no remission group,the positive expression rates were 63.6%,18.2,90.9,63.6%,0.There were statistical differences in the percentage and mean optical density for PTEN 、CD44 between the two groups(p<0.05).The same as the results detected from Image pro plus 6.0 analysis system.Conclusion:High expression of PTEN、CD44 could be an important index for diagnosis and prognosis in Acute Myelogenous Leukemia.
