Observation of Cerebral Microvessels Density in Mice after Chronic Cerebral Hypoperfusion

目的:观察慢性脑缺血小鼠脑内微血管密度变化。方法:(1)KM小鼠12只,随机分为缺血组与假手术组各6只,左侧颈总动脉永久性结扎结合右侧颈总动脉短暂结扎方法制备慢性脑缺血模型,缺血7天后进行脑片TTC染色;(2)TIE2-GFP转基因小鼠12只,随机分为缺血组与假手术组各6只,脑缺血7天后,断头取脑,冰冻切片,荧光显微镜下计数各组小鼠左右双侧皮层、基底节、胼胝体等脑区微血管密度。结果:(1)脑切片TTC染色:与假手术组相比,缺血组有3只小鼠脑切片TTC染色呈淡染;(2)缺血组与假手术组之间左侧皮层、左侧基底节、左侧胼胝体、右侧皮层、右侧基底节、右侧胼胝体等各脑区微血管密度均无显著差异(P均>0.05)。缺血组左右两侧皮层、左右两侧基底节、左右两侧胼胝体微血管密度比较均无显著差异(P均>0.05)。结论:本实验条件下,小鼠脑缺血7天后大脑皮层、基底节、胼胝体等脑区微血管密度未见显著变化。

o observe the changes of microvessel density in mice after chronic cerebral hypoperfusion. Methods: (1) 12 KM mice randomized into two groups: ischemic group and control group. The operation of left common carotid artery ligation and transient right common carotid artery ligation was only applied to ischemic group. 2,3,5-triphenyltetrazolium chloride staining was performed to evaluate infarct volume of brain 7 days after brain ischemia. (2) 12 TIE2-GFP transgenic mice randomized into two groups: ischemic group and control group. The same operation was applied to ischemic group. Frozen sections were prepared to count microvessel density with fluorescence microscope at cortex, basal ganglia, and corpus callosum of right and left sides 7 days after brain ischemia. Results: (1) Brain TTC staining: There are 3 mice`s brain slices lightly stained in ischemic group , compared with control group. (2) Microvessel density comparisons of left cortex, left basal ganglia, left corpus callosum, right cortex, right basal ganglia, right corpus callosum between ischemic group and control group show no significant differences (all P>o.o5) . Microvessel density comparisons between right and left sides of cortex, basal ganglia , corpus callosum of ischemic group show no significant differences (all P>o.o5). Conclusion: The microvessel density of cortex, basal ganglia, corpus callosum show no significant changes 7 days after brain ischemia under current experimental circumstances.
