BRCA1/2是分别在1990年和1994年被发现的遗传性乳腺癌和卵巢癌综合征(Hereditary breast and ovarian cancer (HBOC) syndrome)的易感基因。BRCA1和BRCA2均可维护基因组的稳定性,但二者分别作用于DNA损伤应答和DNA修复的不同环节,前者既可激活细胞周期检测点也对于DNA修复十分关键,而后者则被证明是DNA同源重组修复的一个核心因子。本文将总结近年来的研究成果,阐述BRCA1/2在DNA损伤应答和修复的各个环节中的角色和功能。

BRCA1 and BRCA2 were defined as breast cancer susceptibility genes respectively in 1990 and 1994, since they are mutated in hereditary breast and ovarian cancer syndrome. BRCA1 and BRCA2 suppress tumorigenesis through maintaining the genome stability.However, the two proteins have different roles in the DNA damage response and in DNA repair. BRCA1 functions in both cell cycle checkpoint activation and DNA repair, whereas BRCA2 is a core mediator of homologous recombination (HR).In this review, we focus more on the role of BRCA1/2 in the DNA damage response and in DNA repair.
