采用正交实验对化学-酶协同水解牛毛制备抗氧化肽的工艺进行优化。根据国家标准规定方法,测定牛毛样品的基本化学成分;在单因素实验的基础上,选取酶用量、底物浓度、水解温度和水解时间为考察因素,以水解产物的ABTS自由基清除能力为评价指标对制备工艺进行正交优化。结果表明,牛毛样品含水分11.19 ± 0.21%、灰分6.43 ± 1.71%,粗脂肪2.56 ± 0.60%,蛋白质85.57 ± 5.61%;最佳水解工艺参数为:碱性蛋白酶Alcalase用量5%、底物浓度5%、水解温度60 C、水解时间8h。

Orthogonal design method was used to optimize the chemical-enzymatic hydrolysis of bovine hair to prepare antioxidant polypeptides. With the analysis using National Standard of People's Republic of China, bovine hair was observed to be composed by water (11.19 ± 0.21%), ash (6.43 ± 1.71%), crude fat (2.56 ± 0.60%) and protein (85.57 ± 5.61%). According to the results of single-factor test and orthogonal design method, the optimazed parameters for bovine hair hydrolysis were concluded as: enzyme dosage (5%), substrate concentration (5%), temperature (60 C) and time (8h).
