高温致神经管畸形相关基因Cx43的克隆及其致畸相关性研究

目的 通过筛选高温致畸金黄地鼠胚胎神经管cDNA文库,克隆高温致畸相关基因Cx43,并探讨其与高温致NTDs发生之间的关系。方法 以Cx43特异性引物经PCR方法,利用96孔板逐级筛选高温致畸金黄地鼠胚胎神经管cDNA文库获取阳性克隆。将得到的阳性噬菌体进行质粒转化、酶切鉴定及测序验证。回收基因片段,标记为探针,与孕8.5d高温致畸胚胎神经管组织和正常对照胚胎神经管组织总RNA进行Northern杂交。 结果 经五轮筛选得到含长度为1304bp的金黄地鼠Cx43cDNA片段的阳性噬菌体,其序列与gene bank中金黄地鼠Cx43的序列同源性一致。Northern杂交结果显示其高温致畸组的表达则明显高于正常对照组。 结论 高温未引起Cx43基因变化,但其在神经管中的表达增高与高温致NTDs的发生密切相关。
Objective To screen connexin 43 gene from the cDNA library of neural tube of golden hamster treated with hyperthermia. Discuss the relationship between connexin 43 gene and the NTDs caused by hyperthermia. Methods Based on the PCR method, 96-hole cell culture plate was used to screening the cDNA library of neural tube of golden hamster treated with hyperthermia. Using a pair of Cx43 specific primers,the positive hole was located using PCR. After selective amplification the positive plaques were identified. Then positive plaques were converted to plasmids, enzyme identified, and the sequence was analyzed to confirm the result. The cDNA fragment was extracted and labeled by DIG with random priming method. Then the probes were used to hybrid with total RNA of neural tube of heat-treated groups or control groups at E8.5d to affirm its abnormal expression in hyperthermia induced NTDs. Results The positive clone containing Cx43 cDNA fragment was obtained after five screenings. Results showed that the length of the cDNA insert was 1304bp and its sequence had the same homology with the CX43 of golden hamster in the gene bank. Northern blot showed that its expression in heat-treated groups was evidently higher than control groups. Conclusion Hyperthermia could not induce any mutations in Cx43 cDNA. The increasing expression of Cx43 is intimate with neural tube defects induced by hyperthermia.
