目的:初步探索RNA m6A甲基化阅读蛋白YTHDF2在食管鳞癌放射敏感性调控中的作用。方法:采用重组腺病毒载体与siRNA分别上调/下调人食管鳞癌细胞系TE-1和KYSE-30中YTHDF2表达,通过细胞克隆形成、细胞凋亡、DNA损伤、活性氧水平检测方法,探讨YTHDF2对食管鳞癌细胞放射敏感性的作用与机制。结果:经4 Gy X射线辐照后,TE-1细胞中m6A总水平在24h内先升高后降低;YTHDF2的mRNA水平、蛋白水平也在24h内呈现剂量-时间依赖的先升后降趋势。与对照组相比,过表达YTHDF2后TE-1细胞受照后克隆形成能力显著增强(TE-1: SER = 0.887222)、细胞凋亡率下降(TE-1: P = 0.0052)、活性氧水平显著下降(TE-1: P = 0.0117)、DNA双链断裂的marker γ-H2AX表达量减少。反之亦然。结论:RNA m6A甲基化阅读蛋白YTHDF2参与了对食管鳞癌放射敏感性的调控作用,其作用机制可能与其参与调控食管鳞癌细胞中由辐射诱导的凋亡、活性氧生成及DNA损伤有关,但所涉及的具体分子机制仍有待进一步深入研究。

Objective: To primarily explore the roles and mechanisms of YTHDF2, the RNA m6A methylation reader protein, in regulating the radiosensitivity of esophageal squamous carcinoma (ESCC) cells. Methods: Recombinant adenovirus and siRNA were used to up/down-regulate YTHDF2 expression in human ESCC cell line TE-1 and KYSE-30, respectively. The effects and mechanisms of YTHDF2 on radiosensitivity of ESCC cells were investigated by colonogenic, apoptosis, DNA damage, and ROS assays. Results: After exposed to 4 Gy X-ray irradiation, the total m6A level in TE-1 cells was determined. And it was showed that total m6A in TE-1 was increased and then decreased within 24h. The mRNA level and protein level of YTHDF2 also showed a similar tendency. Compared with the control group, after overexpression of YTHDF2, the clone formation ability of TE-1 was obviously enhanced (TE-1:SER=0.887222) and the apoptosis rates were decreased (TE-1:P=0.0052), the level of ROS was decreased (TE-1:P=0.0117), and the DNA damage was reduced. And vice versa. Conclusion: The RNA m6A methylated reader protein YTHDF2 was involved in the regulation of radiosensitivity in ESCC cells. The mechanism might be related to its involvement in the regulation of radiation-induced apoptosis, ROS generation and DNA damage in ESCC cells. However, the specific molecular mechanisms involved were remain to be further investigated in depth.
