大豆花叶病毒病(SMV)是全球主要大豆产区分布最广的病毒性病害之一,严重影响大豆的产量和品质。本研究在前人研究的基础上,利用对SMV N3株系具有成株和种粒斑驳抗性的抗源东农93046与感病品种品1246构建重组自交系群体,以期探明抗源东农93046对SMV N3株系成株和种粒斑驳抗性的遗传方式,同时利用SSR标记解析东农93046的成株和种粒斑驳抗性位点并获得用于辅助选择的分子标记。结果表明:抗源东93046对SMV N3株系的成株和种粒斑驳抗性均为质量性状,其遗传均受单基因控制;利用前期筛选获得的13个普通SSR标记和新开发的22个SSR标记构建了一个包含35个SSR标记、总遗传距离为260.1cM、平均遗传距离为7.44 cM的F连锁群分子遗传连锁图谱,并获得了对大豆花叶病毒病N3株系成株抗性及种粒斑驳抗性均紧密连锁的SSR分子标记Satt114、Satt334、Satt362,二者具有相似的染色体位置;同时验证了紧密连锁标记具有较高的分子标记辅助选择的准确性,为分子标记辅助选择奠定了良好的基础。

Soybean mosaic virus (SMV) is a prevalent virus,which seriously impacts on soybean yield and quality. Based on previous study, one recombinant intercross line (RIL) population from a cross between Donong93046(with resistance to plant and seed coat mottle of SMV N3) and Pin1246(with susceptible to plant and seed coat mottle of SMV N3) were used to analyze plant and seed coat mottle resistance inheritance mode of Donong93046, to map SMV resistant gene loci through SSR markers, and to obatain SSR markers assoiated with resistant gene loci for marker assistanat breeding. The result of the inheritance mechanism of plant and seed coat mottle resistance in Donong93046 was both quality trait, which were both contolled by a single resistant gene. A molecular genetic map of F linkage group was consructed based on 13 original SSR markers and 22 new developed SSR markers, which covered 260.1cM (with mean marker distance:7.44 cM). The plant and seed coat mottle resistance gene loci were both associated with SSR marker Satt114, Satt334 and Satt362, which were located in similar genomic regions. Furthermore, the coincidence ratio of associated SSR marker in phenotype and genotye of line were very high, which could be used to marker assistant breeding.
