蚊媒体病是全世界重要的传染病之一,严重危害人类健康。化学防治因其速杀、长效、方便等特点,一直是蚊媒综合治理策略中的主要方法。然而随着杀虫剂连续、不合理的使用,蚊媒抗药性也随之开始产生和发展。蚊媒抗药性产生和发展加剧了蚊媒病传播的恶化趋势。化学防治在以后很长的时间内仍是蚊媒治理的主要方法。因此蚊媒抗药性研究仍是控制蚊媒病传播的重点。蚊媒抗药性机制包括3个主要方面:一是靶标抗性,即蚊媒杀虫剂靶位点的改变;二是代谢抗性,其涉及代谢相关酶类转录水平的变化;此外表皮抗性也发挥重要作用,如表皮改变影响杀虫剂的穿透性。 Piwi-interacting RNAs (piRNAs) 是2006年几个实验室发现的一类新的非编码小RNA(sncRNA)。本研究报告了淡色库蚊抗药性相关piRNAs表达谱。高通量测序技术在敏感品系(DS)和抗性品系(DR)中获得了112608个piRNA clean reads,其中44.8%clean reads可比对致倦库蚊的piRNAs。DS得到515198 reads,DR得到222160 reads;序列去冗余后的piRNA类别数目,在 DS 中有40456个,DR有15519个;重复序列中,DS匹配的piRNA有20343个,DR匹配的piRNA有10889个,混合样本匹配的piRNA是16090个,匹配到的piRNA在DS 、DR和混合样本中均没有倾向性;基因组统计的结果相似;piRNA分子广泛分布在基因间;piRNA长度在28nt和31nt出呈现出两个峰值;此次测序共获得差异表达的piRNA分子50445个,DS高表达37608个,DS低表达12837个。
he mosquito-borne diseases are important infectious diseases in the world and caused serious threat to human health. Chemical control, because of its killing speed, long-lasting and convenient features, has been the main method in the comprehensive management strategy. Unfortunately, unreasonable and continuous usage of insecticides induced the development and spread of resistance. Then insecticide resistance exacerbated the spread of mosquito-borne diseases. However, after a very long time, chemical control for its rapid and efficient advantage will still be the main method in control of mosquito-borne diseases.Insecticide resistance mechanisms include three main areas: target resistance, that is, the change of the insecticide target sites. Metabolic resistance, that is, the change of metabolic enzymes transcription level involved in metabolic resistance, and physiological resistance and behavior avoidance also involved in pestcide resistance, such as cuticle protein and chitin enzyme also involved in insecticide resistance.Piwi-interacting RNAs ( piRNAs ), 26-31nt, are a new class small non-coding RNA (sncRNA) found in 2006 in several laboratory.In this research,piRNA expression profiling of pestcide resistence in Culex pipiens pallensmosequito.112608 piRNA unique clean reads were gotten using Illumina Hiseq 2000 technology, and 50445(44.8%)clean reads mapping to Cx. pipiens quinquefasciatus.The unique piRNA typle number of DS and DR strain is 40456 and 15519 respectively. We got 515198 piRNA reads in DS and the number in DR is 222160. Meanwhile, the aligned piRNA number in repetitive sequence of DS, DR and pooled sample are 20343, 10889 and 16090 respectively. Matched piRNA show no tendency in DS,DR and mixed sample. PiRNAs from the genome show the similar result. piRNAs widespread express in genes, no matter where they are. Those mapping piRNA show two peaks in 28nt and 31nt in length.The differently expressed piRNAs is50445, the number high express in DS, DR is 37608,12837 respectively.
