人生长激素类似蛋白(human Growth Hormone-like,hGH-L)是首次在五龄蚕血淋巴和蚕蛹中发现的一种在天然状态下能与人生长激素抗体结合的蛋白,分子量为30kDa。本研究以新鲜蚕蛹为原料,经过低温匀浆、过滤、离心去除大颗粒杂质,上清用60%硫酸铵沉淀,再经PBS重悬,通过分子筛脱去色素并进行初步分离,亲和层析纯化hGH-L蛋白。双向电泳后进行MALDI-TOF-MS 质谱分析,氨基酸序列比对后发现,hGH-L蛋白由两个同属β-trefoil超家族的两个脂蛋白组成,其β结构域与hGH空间结构类似。通过k562细胞体外检测其生物活性时发现该蛋白可促进细胞的分裂,为了进一步验证其类促生长作用,从蚕蛹cDNA中克隆到了hGH-L其中的hGH-L2的基因,构建重组家蚕杆状病毒BmNPV-hGH-L2,以低滴度感染家蚕细胞BmN,经流式分析证明该蛋白能缩短细胞G0/G1期,快速通过G1/S转换点进入S期,促进细胞分裂。hGH-L蛋白的发现,将再次引起人们对家蚕药用价值的关注,为新型激素类药物的开发奠定基础。

Human Growth Hormone-like(hGH-L)was first found in the hmolymph of fifthinstar larvae of the Bombyx mori and silkworm pupa which can bind with the antibody of hGH protein in the natural state, its weight is about 30kDa. In this research, we use fresh silkworm pupa as material, discard impurity substance with homogenate, filtration and high speed centrifuge, then collect the supernatant and remove the grease by ammonium sulphate precipitation, resuspend precipitation, depigment and further isolated with gel filtration chromatography. Finally, hGH-L protien was extracted by affinity chromatography. we analysised these protein with 2D electrophoresis, digged the interested protein dots from 2D PAGE and then digested with trypsin. The protein's PMF (Peptide Mass Fingerprinting) was obtained through the MALDI-TOF-MS. At last, we found hGH-L protien consists of two lipoproteins which belong to β-trefoil superfamily and their β-strcuture domains have a close affinity to the structure of hGH. It is indicated that hGH-L protien can promote k562 cells division by in vitro biological assay. In order to further validate the growth-promoting effect of hGH-L, we clone the gene sequence of hGH-L2 from cDNA silkworm pupa, construct the recombinant BmNPV and infect BmN cells as low titer. Flow cytometry assay indicated that hGH-L protein results in shortening of G0/G1 phase, switching cell to G2 phase through G1/S check point quickly and accelerating cell cycle progression and cell proliferation. Beacuse of discovery of hGH-L protein, people will focus on the medicinal value of silkworm again, which will set up a substantial foundation for the developing of novel hormone drugs.
