茶叶中EGCG防治大鼠非酒精性脂肪肝的实验研究
Research on Therapeutic Effect of EGCG from Tea against Non-alcoholic Fatty Liver Disease in Rats
以高脂饲料喂养大鼠,建立非酒精性脂肪肝大鼠模型;分别以低剂量(10 mg/kgoBW)、中剂量(50 mg/kgoBW)、高剂量(100 mg/kgoBW)表没食子儿茶素没食子酸酯(EGCG)灌胃大鼠,探讨EGCG对大鼠非酒精性脂肪肝的防治作用。结果表明, 与模型组相比,EGCG能显著降低非酒精性脂肪肝大鼠的体重,降低血清中甘油三酯(TG)、总胆固醇(TC)和低密度脂蛋白-胆固醇(LDL-C)含量,降低丙氨酸氨基转移酶(ALT)和天冬氨酸转氨酶(AST)活性水平,提高高密度脂蛋白-胆固醇(HDL-C)的含量,改善大鼠肝脏氧化应激状态。肝脏切片显示,EGCG能减轻肝脏细胞中脂滴的形成,对非酒精性脂肪肝大鼠肝脂肪变性具有较强的修复作用。脂质代谢相关基因表达分析显示,EGCG能够调理脂质代谢。实验结果表明EGCG能有效改善和修复大鼠非酒精性脂肪肝损伤。
In order to investigate the therapeutic effects of EGCG on the non-alcoholic fatty liver disease (NALFD), experimental groups were given daily oral administration EGCG with low dose of 10 mg/kgoBW, middle dose of 50 mg/kgoBW and high dose of 100 mg/kgoBW, respectively. The results showed that EGCG could lower the body weight gains of NAFLD rats, reduce the concentration of triglyceride (TG), total cholesterol (TC), low density lipoprotein-cholesterol (LDL-C), activity of alanine aminotransferase (ALT) and spartate aminotransferase (AST), increase the concentration of high density lipoprotein-cholesterol (HDL-C) in the serum, and ameliorate liver oxidative stress compared with the model group. Histological analysis showed that EGCG could reduce the formation of lipid droplets in the liver cell, strongly repair adipohepatic degree of NALFD rats. RT-PCR showed that EGCG could regulate the fatty metabolism genes. The experiment result indicated that EGCG effectively improve and recovery liver injury of NALFD rats induced by a high fat diet.
