家蚕Y-Box蛋白的克隆表达以及功能分析

Y-box蛋白具有高度保守的结构域csd(cold shock domain),是真核生物中的冷休克蛋白。我们通过RT-PCR的方法从家蚕蛹中扩增到家蚕Y-box蛋白基因的完整的ORF序列,并将其命名为Bombyx Y-box(BYB)基因,GenBank登录号为AB098537.1。通过将扩增到的BYB基因克隆到融合表达载体pET-28a相应的酶切位点上,得到重组表达质粒pET-28a-BYB,将该重组载体转化大肠杆菌BL21(DE3),诱导表达后,以亲和层析的方法纯化目的蛋白His-BYB。将纯化的重组蛋白免疫新西兰大白兔制备多克隆抗体, ELISA检测效价大于1∶12800。采用实时荧光定量PCR方法和免疫印迹方法(Western blotting)对家4个发育时期以及5龄幼虫各组织中BYB的mRNA转录水平和BYB蛋白进行检测。应用免疫细胞化学技术对家蚕Bm5细胞进行亚细胞定位分析,发现BYB蛋白在整个细胞周期主要分布于细胞质中,但在4℃冷激2h后,BYB蛋白则同时位于细胞核和细胞质内。推测BYB蛋白在冷激情况下向细胞核内迁移,并对某基因产生调控作用,从而表达相应的应激性蛋白。
he Y-box proteins are a family of highly conserved nucleic acid binding proteins which exist widely in various organisms from bacteria to human.The full open reading frame (ORF) of Bombyx mori Y-box was obtained from silkworm pupa by RT-PCR and named BYB (Bombyx mori Y-box), of which the accession number was AB098537.1 After cloned into pET-28a and expressed in E.coli BL21 (DE3), the BYB was fused with His-tag and purified on chelating columns. The purified fusion protein was used to immunize a male New Zealand rabbit. Then, antiserum was harvested, and IgG was prepared by affinity chromatography using immobilized protein A.BYB in some tissues of the fifth instar larvae, such as head, Malpighian tubule, epidermis, silk glands, seminal glands, gut and fatty body, were extracted and analyzed by Western blotting. The result showed that BYB existed in larva’s seminal glands,Epidermis, and so on. And the similar results was obtained when we applied immunohistochemistry analysis to the fifth instar larva and pupa. We also compared the quantity of BYB mRNA in different tissues of the fifth instar larva and different stages of silkworm growth. These results suggested that BYB was selectively expressed in silkworm’s seminal glands, epidermis and Malpighian tubule. After subcellular localization with BYB, we found that it mainly existed in cytoplasm under normal conditions.However, after Bm5 cell had been treated with cold shock of 4℃ for 2 h, most of the BYB protein distributed in the cytoplasm and nucleus. This research laid a good foundation of further studies on BYB.
