骨胶原多肽对小鼠抗氧化能力和皮肤、骨骼胶原蛋白mRNA表达的影响
EFFECT OF OSSEIN PEPTIDE ON ANTIOXIDANT CAPACITY AND COLLAGEN mRNA EXPRESSION OF SKIN AND BONE IN HIGH-FAT-DIET KM MICE
目的:研究骨胶原肽(OP)对长期高脂膳食小鼠抗氧化能力和皮肤及骨骼中胶原表达的影响。 方法:75只KM小鼠分成5组,分别是正常组,高脂组,硫辛酸(LA)组,1%OP组和2%OP组,饲养16W,眼球采血宰杀小鼠,取约100 mg皮肤用于抗氧化能力测定,再取约100 mg皮肤和左侧股骨头用于胶原蛋白基因表达的测定。 结果:高脂饲料可导致小鼠抗氧化能力减弱,皮肤和骨骼中胶原蛋白表达量显著下调(P<0.05),而添加了OP可显著降低其ROS水平及脂质过氧化物含量(P<0.05),显著提高过氧化氢酶活力和总抗氧化能力,还能有效上调组织中主要胶原蛋白的表达量(P<0.05),高剂量OP能使大部分指标恢复到正常水平,LA也有一定效果,但相对较弱。 结论:添加OP对高脂日粮小鼠的抗氧化能力和胶原蛋白mRNA表达量均有显著的提高作用,并且证明了机体氧化还原状态与胶原代谢之间密切相关。
Purpose] To explore the influence of ossein peptide (OP) on the antioxidant capacity and collagen mRNA expression of skin and bone in high-fat-diet km mice. [Methods] 75 KM mice were randomly assigned to 5 groups, which respectively control group, high-fat-diet group, lipoic acid (LA) group, 1%OP group and 2%OP group. All mice were sacrificed at the end of 16ws, 100 mg skin were taken for the measurement of antioxidant capacity, 100 mg skin and heads of left femurs were taken for collagen genes expression. [Results] High-fat-diet could weaken antioxidant capacity, decrease collagen genes expression in skin and femur. In contrast, OP could decrease the ROS level and MDA content significantly (P<0.05), strengthen the activity of CAT, T-AOC and up-regulated the main kinds of collagen mRNA expression in skin and femur (P<0.05) significantly. High does OP can recover most of the indicators to normal level, LA also worked, but the effects were not as good as OP. [Conclusion] The supplements of OP could improve antioxidant capacity and collagen mRNA expression,and the close relationship between the body's redox status and collagen metabolism was also be confirmed.
